PURPOSE:Many researchers claim that Age-related maculopathy susceptibility 2 gen(eARMS2) is associated with age related macular degeneration(AMD), especially advanced AMD. But its role in the disease is not clear. Through our basic experimental study to ARMS2,to figure out the genetic factors in disease occurrence, development process, and prevention of disease.Methods1.Five human RPE eyes for this study were isolated from donors. We cultured primary human retinal pigment epithelial (RPE ) cells which were taken from the macula and peripheral retina to compare . The samples from the same eye are divided into the same group.2.Semiquantitative reverse transcription-polymerase chain reaction was used to study ARMS2 gene expression.3. Western blot analysis were used to study ARMS2 gene and protein expressionResultThe levels of ARMS2 mRNA in cells cultured from macula are than those in cells grown in ellipsoid region(P<0.05). While the levels of ARMS2 protein in cells cultured from macula are two times higher than those in cells grown in ellipsoid region. (P<0.05) .ConclusionThe ARMS2 expressions of macula are much more than that of the peripheral retina. This study represents that ARMS2 may play an important role in the pathology,occurrence, development process of AMD.
