Objective To test and analysis the urinary metabolites of autistic children and normal children , research for specific metabolites in the urine of autistic children, establish indicators of urinary metabolites in autistic children,provide experimental evidence for the early screening and diagnosis of children with autism.Methods Applied nuclear magnetic resonance spectroscopy and pattern recognition, the urine composition of the two groups,which were autism group and control group, was analysised and comparised. In the autism group the relationship of special urine metabolites and symptom severity was explored.Resultsâ‘ . 1HNMR spectra of urine showed that the urinary metabolites: N-methyl-2-pyridone-5-formamide(2PY), N-methyl -nicotinic acid(NMND), and Hippurate were significantly different between autism group and control group (tNMND=7.594, P=0.000<0.05, t2PY=70.042, P=0.000<0.05, tHippurate= -17.461, P=0.000<0.05).â‘¡. The concentration of NMND and 2PY in children with autism and the ABC scale score had a positive correlation(P=0.000,P<0.05), the concentration of Hippurate in children with autism and the ABC scale score had negitive correlation(P=0.000 , P<0.05).â‘¢. The concentration ratio of specific urinary metabolites and creatinine(NMND,2PY and Hippurate) was a significant difference between autism group and control group(P=0.000,P<0.05).Conclusionâ‘ In the autism group,the concentrations of 2PY and NMND both increased,while the concentrations of Hippurate reducted.â‘¡There is a relevantion between ABC scale score and urinary metabolite concentrations in autism group. The concentration of NMND and 2PY in children with autism and the ABC scale score has a positive correlation, the concentration of Hippurate in children with autism and the ABC scale score has negitive correlation,which indictated NMND,2PY and Hippurate were screening indicators of urine metabolomics for Children with autism.This conclusion can be confirm by expanding the sample ,analysising specific urinary metabolites and establishing animal model of autism.
