Ramulus mori., as a commonly used traditional Chinese medicine, the current researches on its components and quality control study are limited.So it is significant to use new separation and analytical techniques for comprehensive research of Ramulus mori..In our study, preparative high performance liquid chromatography and high speed counter current chromatography were successfully applied for separation from Ramulus mori..After optimum conditions were used,five high purity components were obtained:mulberroside A,mulberroside F, trans-oxyresveratrol, trans-resveratrol and moracin M,which were confirmed by MS and NMR, mulberroside F was isolated for the first time from Ramulus mori.In additon, an accurate, reliable and reproducible method of high performance liquid chromatography coupled with photodiode array detector(HPLC-DAD) both for simultaneous determination of five components and fingerprint was developed for the first time.And in combination with a suite of chemometrics methods as similarity analysis(SA),principal component analysis(PCA) and hierarchical clustering analysis(HCA), the data of 25 batches of Ramulus mori.from different origins were evaluated statisticlly. It was found that the content of each analyte and the similarity varied greatly among the different samples, it indicated that the qualities of 25 battches Ramulus mori.were different.And it was hard to classify the samples and identify the key categorizing parameters with their origins in PCA and HCA plot. It is concluded that the origin may not the important factor influencing on the quality of Ramulus mori.,maybe the the harvest time or processing method have some relationship with it.The study could provide an experimental basis for development of its quality control,and it would be an important referrence for utilization of Ramulus mori. resources.
