| Objective:To investigate part mechanism of invigorating the kidney and activating blood circulation compound recipe Yizhi Jiannao granule Concentration Fluid (YCF) on preventing and curing Alzheimer's disease by observing the effect of YCF on AD rats'learning and memory ability, the expression of insulin receptor (IR) mRNA and glucose transporter 4(GLUT4) mRNA in the hippocampus of AD rats.Methods:40 male AD rats were randomly divided into 4 groups, the normal group(group A),sham operation group (group B),model group(group C), and the YCF group (group D). Then Aβ25-35 were injected in both sides of hippocampus of the last two groups to make AD models.The same volume of Normal saline were injected in hippocampus of group B Rats in the same way. No handling to group A.All the groups were tested with Morris water maze test (MWM) after operation to test whether AD models were successful or not..Rats of group D were given intragastric administration with yizhi jiannao granule concentration fluid (YCF), while the other 3 groups were given double-distilled water for 8 weeks. All the groups were tested with Morris water maze test (MWM) again after above treatment to observe the change of the all rats'learning and memory ability.Finally, the hippocampus were taken out to detect the expression of IR,GLUT4 mRNA with RT-PCR. Results:1.Behavioral results:In the hidden platform searching and exploratory experiment, the group C and D rats' learning and memory ability delayed significantly (the escaping latent period is prolonged, the time staying in the original platform and the average frequency of passing through hidden platform is decreased) compared with group A after Modelling.After traditional Chinese medicine intervention, The group D rats' learning and memory ability were significantly strengthened (the escaping latent period is delayed, the time staying in the original platform and the average frequency of passing through hidden platform is increased) compared with group C.2. RT-PCR results:The expression of IR and GLUT4 mRNA in group C decreased significantly (P<0.01) Compared with group A; there is no significant difference between group B and group A (P>0.05)The expression of IR,GLUT4 mRNA in group D increased significantly (P<0.01) Compared with group C.Conclusion:1. By injecting condensed Aβ25-35 to both sides of rats' hippocampus can make representative AD rats model.2. Invigorating the kidney and activating blood circulation compound recipe Yizhi Jiannao granule can remarkably improve AD rats'learning and memory ability.3. Maybe the expression of IR and GLUT4 mRNA in the AD rats' hippocampus were delayed is one of the pathogenic mechanisms of Alzheimer's disease.4. Yizhi Jiannao granule maybe treat AD by increasing the expression of IR, GLUT4 mRNA in AD rats' hippocampus,thereby improving insulin resistance, reducing the form of Senile Plaques and neurofibrillary tangles, protecting the neurons. |
