To investigate the relationship of C-reactive protein polymorphisms (+1059G>rs1800947,+1444C>T rs1130864) and haplotypes with the susceptible possibility of chronic periodontitis and/or coronary heart disease; (2) To investigate the relationship of periodontal pathogens between chronic periodontitis and coronary heart disease Methods:(1). DNA was obtained from buccal swabs of 100 patients with moderate to severe chronic periodontitis (CP group),98 patients with coronary heart disease (CHD group),92 patients with moderate to severe chronic periodontitis with coronary heart disease (CP+CHD group) and 125 matched healthy controls (control group). Genetic polymorphisms of+1059G>C rs1800947 and+1444C>T rs1130864 were analyzed with PCR-PFLP technique and the genetic haplotypes were identified by sequencing in CP+CHD group;(2) DNA was obtained from coronary atherosclerotic plaque and subgingival plaque of 38 cases that were randomly selected from CP+CHD group,then to detect Fusobacterium nucleatum (Fn) and Tannerella forsythensis (Tf) in the obtained DNA by RTQ-PCR. Results:(1) There was no significant difference in+1059G >C rs1800947 and+1444C>T rs1130864 distribution of genotype among the four groups; (2) The difference in+1444C>T rs1130864 polymorphism had the tendency of expanding in CHD+CP group. (3)There was a difference in haplotype GT of rs1800947 and rs1130864 between control group and CP+CHD group. (4)subgingival plaque:Fn/ total bacteria=1.68% Tf/total bacteria=0.842%. Atherosclerotic Plaque:Fn/total bacteria=0.313%. Tf/total bacteria=0.221%. (5)The detection rate of periodontal pathogens in subgingival plaque and coronary atherosclerotic plaque:Fn (93.3..0%,86. 0%),Tf (86.7%,30.0%). (6)The detection of Fn was relevant with Tf in subgingival plaque (P=0.014,r=0.681) but there was no relevance in coronary atherosclerotic plaques (P=0.563) Conclusion:(1) The allele of haplotype GT is the predisposing gene of coronary heart disease and chronic periodontitis.
