| ObjectiveHair dye dermatitis is a commom allergy contact dermatitis in clinics, and para-phenylenediamine is one of the main allergens. The mechanism of PPD induced allergy dermatitis may be associated with environmental exposure factors, individual immunity and genetic susceptibility. The purpose of this study is to explore the relationship between genetic polymorphism of NAT1, NAT2 and ACE and susceptibility to hair dye dermatitis, by investigating the distribution of NAT1, NAT2 and ACE genotype between the patients with hair dye dermatitis and the healthy individuals.MethodsCollecting blood samples of 60 hair dye dermatitis patients and 73 control subjects from February,2009 to February,2010 and extracting DNA from the blood samples. The distribution of alleles and genotype of NAT1, NAT2 and ACE were detected by PCR-RFLP technique.The association between the three genes genotype and susceptibility to hair dye dernatitis was analyzed.Results1. NAT1 gene:The frequency of NAT1*10 in hair dye dermatitis group was 35.8%, significantly lower than the control group (χ2=3.954, P<0.05); In hair dye dermatitis cases, the frequency of NAT1*4/*4, NAT1*4/*10, NAT1*10/*10, NAT1*3/*10 and NAT1*3/*4 were 36.7%,40.0%,15.0%,1.7% and 6.7%, respectively, no statistically significant difference of the frequencies was found between the two groups. In hair dye dermatitis cases, the frequency of rapid phenotype NAT1 was 56.7%, which was slightly lower compared with the control group (71.2%,χ2=3.058, P=0.08).2. NAT2 gene:In hair dye dermatitis cases, the frequency of NAT2*4, NAT2*5A, NAT2*6B, NAT2*7A was 52.5%,5.0%,26.7% and 15.8%, respectively, no statistically significant difference of the frequencies was found between the hair dye dermatitis patients and controls. The frequency of rapid genotype, intermediate genotype and slow genotype was 26.7%,51.7% and 21.7% in hair dye dermatitis cases,30.1%,50.7% and 19.2% in control subjects, no statistically significant difference of the frequencies between the two groups. The frequency of rapid phenotype and slow phenotype was 78.3%,21.7% in hair dye dermatitis cases, no statistically significant difference of the frequencies between the two groups.3. ACE gene:In hair dye dermatitis cases, the frequency of I and D allele was 55.8% and 44.2%, respectively, no statistically significant difference of the frequencies was found between the hair dye dermatitis patients and controls; In hair dye dermatitis cases, the frequency of II genotype was 33.3%, significantly higher than the control group (χ2=4.255, P<0.05).Conclusion1. In hair dye dermatitis cases, the frequency of NAT1*10 was significantly lower than the control group, the frequency of rapid phenotype was slightly lower than the control group, no significant difference of genotype between two groups.2. No statistically significant difference of the frequencies of NAT2 allele, genotype and phenoty between hair dye dermatitis cases and controls.3. In hair dye dermatitis cases, the frequency of ACEⅡgenotype was significantly higher than the control group, no significant difference of I and D allele between two groups. |
