ObjectiveERCC5 protein and gene mRNA expression level of NSCLC(non-small cell lung cancer)and the cancer adjacent tissues were checked out to comprehend the relationship between the protein expression and the pathological features of NSCLC. The relationship between the ERCC5 protein expression and the survival time after platinum-containing chemotherapy was analysed through follow-up to provide a better theoretical basis and clinical-pathological characteristics for further screening NSCLC prognostic factors and individualized treatment.Method1,The level of ERCC5 protein expression in 86 NSCLC and adjacent cancer tissue got from the patients who had thoracic surgery in Liaoning Medical Coll -ege from February 2002 to February 2006 was checked out by immunohi- sto chemical method to analyse the expressive feature of different types in NSCLC and its role occurrence and development.2,The level of ERCC5 mRNA expression in 30 NSCLC and adjacent cancer tissue got from the patients who had thoracic surgery in Liaoning Medical Coll -ege from February 2010 to November 2010 was measured by RT-PCR meth -od to investigate the relationship between ERCC5 protein expression and gene mRNA expression.3,The relationship between survival(1,3,5 year survival r OS,median TTP ,MST) after platinum-containing chemotherapy and ERCC5 expression was investiga -ted through follow-up study. Results(1) The level of ERCC5 in NSCLC is higher than in cancer tissues.(P<0.05)(2) ERCC5 protein expression in NSCLC with the degree of differentiation were statistically significant. (P<0.05) The lower differentiation has lower positive rate of ERCC5 expression,While, there were no statistical significance of the patient's sex,age,pathological type and lymph node metastasis. (P>0.05) .(3)The level of tissues gene mRNA expression of ERCC5 in NSCLC was significantly higher than the adjacent tissues.There was statistically significant between two groups.(P<0.05).(4) 86 patients with NSCLC surgery alone group (36 cases): ERCC5 protein expression postoperative overall survival was significantly longer than the negative expression group .(P<0.05).(5) 86 patients with NSCLC surgery and received postoperative platinum containing chemotherapy group (50 cases): ERCC5 protein expression postoperative overall survival was no significantly longer than the negative expression group .(P>0.05).Conclusions(1) The level of ERCC5 expression in non-small cell lung cancer is closely related to the degree of differentiation. the level of ERCC5 expression is lower following the degree of differentiation, and it suggested that the ERCC5 expression is related to higher invasiveness and malignant behavior of tumor. It suggested that ERCC5 plays a important role in development of tumor, and ERCC5 may become a factor that predicted the degree of malignancy and prognosis of NSCLC.(2)In NSCLC, the variation of ERCC5 expression may be one of the mechanisms of differential expression of its protein.(3) In NSCLC,ERCC5 may related to platinum-based resistance; ERCC5 may become sensitive factor of non-small cell lung cancer chemotherapy, and to guide individual treatment.
