ObjectiveObjective: To observe the effect of PMMA bone cement particles on the expression of SOCS1and SOCS3 in rat macrophages cultured in vitro.Methods: The purity and activity of the macrophages screened from peritoneal lavage fluid ofrat and cultured in vitro were identified by CD68 monoclonal antibody immunocytochemistry.PMMA bone cement particles were added into the macrophages according to the experimentaldesign, and the phagocytosing of PMMA particles was observed and the expression of SOCS1and SOCS3 were detected by real time fluorescence quantitative PCR (Real-time PCR). Theeffects of PMMA bone cement particles on the expression of SOCS1 and SOCS3 were observedboth in different concentrations of particles and a certain concentration of particles at differenttime.Results: PMMA bone cement particles increased the expression of SOCS1 and SOCS3 ofmacrophages in a time-and dose-dependent fashion. The expression of SOCS1 and SOCS3 inmacrophages were elevated and reached its peak(P<0.05), respectively when the concentration ofPMMA bone cement particles was 0.25mg/ml and cultured with particles for 24h in vitro.Conclusion: PMMA bone cement particles can facilitate the expression of SOCS1 and SOCS3in rat macrophages, which further enhance the inhibition of signal transduction of somecytokines such as IFN-γ,IL-6, and then promote the differentiation of osteoclast precursors andaccelerate the osteolysis. Selective suppression of SOCS1 and SOCS3 in osteoclast precursorsmay be a possible therapeutic strategy for inflammatory bone destruction.
