| Objective 1. Because the different solubility in different urine collection conditions. The collection conditions will influence the outcome. Study the methods of 24-hours urine risk factors preserved, then preserve 24-hours urine risk factors become more easy and precise.2. Preserve 24-hours urine risk factors for a long time was a puzzle, and we have little investigation for that. So we study adding antiputrefactive or preservative which could preserve 24-hours urine risk factors for a long time, then analysis 24-hours urine risk factors become more easy.Patients and methods 1. Spot urines(n=30) were obtained from patients for calciuria, oxaluria,citraturia, phophauria and uricosuria analysis. Then one single spot urine sample was divided into five aliquots of 10 mL each: one containing of HCl (12mol/LHCL,100ul); one containing sodium bicarbonate(10g/L NaHCO3,100ul); one containing Thymol(0.2g); one containing methylbenzene(100ul) and one without any preservative (untreated group). Calciuria,oxaluria,citraturia,phophauria and uricosuria were measured after 24 h sampling in room temperature.2. Spot urine(n=30) were obtained from patients for calciuria,oxaluria,citraturia,phophauria and uricosuria analysis. Then one single spot urine sample was divided into five aliquots of 10 mL each: one containing of HCl (12mol/L,100ul), one containing sodium bicarbonate,( 10g/LNaHCO3,100ul), one containing Thymol(0.2g), calciuria,oxaluria,citraturia,phophauria and uricosuria were measured after 72h sampling in room temperature, and measured again 144h after sampling in room temperature.Result 1.Calciuria, citraturia, phophauria and uricosuria were no significant statistically difference between Thymol group and spot urine group, but oxaluria has statistically difference(P<0.05); calciuria,oxaluria,phophauria and uricosuria were no statically significant between HCL group and spot urine group, but citraturia has statistically difference(P<0.05);calciuria,phophauria and uricosuria were no significant statically difference between methylbenzene group and spot urine group, but oxlaluria and uricosuria have statistically difference(P<0.05); uricosuria were no statically significant between NaHCO3 group and spot urine group; calciuria and uricosuria were no significant statically between untreated group and spot urine group.2. After preserved 72hours, calciuria,citraturia,phophauria and uricosuria were no significant statistically difference between Thymol group and spot urine group, but compare with spot group, oxaluria has decreased; (P<0.05) calciuria,oxaluria,phophauria and uricosuria were no significant statically between HCL group and spot urine group, but citraturia has decomposed(P<0.05); and uricosuria was no difference between NaHCO3 group and spot urine group. after preserved 144 hours, calciuria,citraturia,phophauria and uricosuria were no significant statistically difference between Thymol group and spot urine group too; calciuria,oxaluria,phophauria and uricosuria were no significant statically between HCL group and spot urine group;and uricosuria was no difference between NaHCO3 group and spot urine group.Conclusion 1.We recommend assay of calciuria,oxaluria and phophauria in acidified sample of HCL collection condition, but we cannot assay of citraturia in acidified sample; We recommend assay of calciuria, citraturia, phophauria and uricosuria in sample of thymol collection condition, but we cannot assay of oxaluria in thymol sample; In order to assay all of the 24-hours urine risk factors, we can integrate HCL and thymol.2.Acidified urine can preserve calciuria,oxaluria and phophauria for at least 6 days; and using thymol can preserve calciuria, citraturia, phophauria and uricosuria for at least 6 days, we can integrate HCL and thymol preserving all 24-hours urine risk factors for a long time. |
