ObjectiveTo induce the rat adipose derived stem cells (ADSCs) into smooth-muscle-like cells in vitro.MethodsADSCs were separated and collected from the adipose of inguinal region of SD rats. Growth curve of ADSCs was recorded. The fourth passage ADSCs were used to induce into adipocytes by adipogenic inducing fluid and examined by Oil Red O staining. The fourth passage ADSCs were used to induce into osteoblasts by osteogenic inducing fluid and examined by Von Kossa staining. The fourth passage ADSCs were used to induce into smooth-muscle-like cells by inducing fluid which containβ-mercaptoethanol, and identified by the expression ofα-smooth muscle actin (α-SMA) by immunohistochemistry.ResultADSCs had spindle and polygon shape and could proliferate rapidly in vitro. Growth curve showed the growth of ADSCs turned into logarithmic growth phase two days after subculture. The fourth passage ADSCs observed red stained fats characteristic of adipocytes in the cytoplasm on Oil Red O staining after induction with adipogenic inducing fluid, and observed calcium nodes characteristic on Von Kossa staining after induction with osteogenic inducing fluid. The immunohistochemical result of ADSCs induced into smooth-muscle-like cells indicated the positive rates ofα-SMA inβ-mercaptoethanol induced cells and uninduced cells were (29.80±6.89)%,(2.89±1.24)%. The positive rates in induced ADSCs were remarkably higher than those in uninduced ADSCs (P<0.01).ConclusionADSCs expressed smooth muscle markers after being induced, and they could be a new potential source expected to cure smooth muscle correlative diseases in tissue engineering field.
