Objiective: To observe the estrogen of Soybean isoflavones in different environments, on MMTV-erbB-2 effect of breast cancer, development of transgenic mice.Methods: Select 150 small female rats with MMTV-erbB-2 transgenic and random divided into five groups, random select 60 small female rats that in age of 5 weeks as a rows to operate castration, set for low female hormone group, and random divided into 2 groups, each group 30 only, a group feed feeding beans feed, set for low female hormone Group 1, another group feed feeding controlled feed, set for low female hormone Group 2. random select 60 only small female rats, in age of 5 week Shi up, on small rats for injection female hormone, 50ng/times, 3 times/week, set for high female hormone group, and random divided into 2 groups, a group feed feeding beans feed, set for high female hormone Group 1, another group feed feeding controlled feed, set for high female hormone Group 2. 30 only small female rats set for blank controlled group, feed feeding controlled feed. To observe the small rats of the growth development situation, to execute experimental to the small rats that in age of 60 weeks, with Whole mount dyeing observation breast overall form structure, and immune organization chemical dyeing SP method detection breast organization erbB-2 protein, ER,PR,PCNA of expression, while observation the group small rats breast tumor of incidence and incubation period, and tumor growth parts and tumor growth situation, and analysis its role mechanism.Result: 1:The experimental group tumor occurs rate and the incubation period phase comparison, low female hormone Group 1, low female hormone Group 2, high female hormone Group 1, high female hormone Group 2, controlled group of breast cancer incidence respectively for 96.7% and 30.3%,40%,83.3% and 73.3%, controlled group and high female hormone Group 2, low female hormone Group 1 and high female hormone Group 2, low female hormone Group 2 and high female hormone Group 1 of incidence phase comparison (P>0.05) other the Group phase comparison (P<0.05). Low estrogen environment, soybean isoflavone Group significantly reduced tumor latency than other groups, under the high estrogen environment, soybean isoflavone Group significantly extended the tumor latency than other groups. 2: the expression of the experimental group ER,PR not significantly different, 3: soybean isoflavone in low estrogen on MMTV-erbB-2 under the environment of genetically modified mice occurrence of breast cancer have a catalytic role, in high estrogen environment on transgenic mice MMTV-erbB-2 inhibition of breast cancer. 4: Soybean isoflavones in different environments of estrogen on expression of ER,PR MMTV-erbB-2 of mammary glands of transgenic mice without dependencies.Conclusion: 1, MMTV-erbB-2 transgenic mice grow well in China, with strong reproductive and adaptive capacity. 2, Soybean isoflavones in different estrogen environment on morphology of MMTV-erbB-2 development of mammary glands of transgenic mice have a certain impact. 3, soybean isoflavone in low estrogen on MMTV-erbB-2 under the environment of genetically modified mice occurrence of breast cancer have a catalytic role, in high estrogen environment on transgenic mice MMTV-erbB-2 inhibition of breast cancer. 4, Soybean isoflavones in different environments of estrogen on expression of ER,PR MMTV-erbB-2 of mammary glands of transgenic mice without dependencies.
