| Partâ… To Establish A Rat Model of Polycystic Ovarian SyndromeObjective: To select a more suitable rat model through the comparison of two kinds of methods which could induce the rat model with polycystic ovarian syndrome.Methods: Forty female SD rats aged 6 weeks were divided into control group 1, model group 1, control group 2 and model group 2. Control group 1 received vehicle only 1% CMC once daily orally, while model group 1 was administered letrozole dissolved in 1% CMC once daily, the treatment period was 21 days; Control group 2 was administered with NS per day from day 1 to 10 and twice-daily from day 11~22. Ins was administered once daily on day 1~22 for model group 2, and the human chorionic gonadotropin (HCG)was administered from day 11 to 22 twice-daily. Observe the estrous cycle by vaginal smear, the ovarian augmentation by weight of two ovaries, ovulation and follicle change by ovarian histology and the serum level of hormones by RIA.Results: The vaginal smears from the model group 1 showed that all rats lost steady estrogen cycle and in interphase. Compared to control group 1, the weight of the rats and two ovaries were increasing, the ovarian showed multiple follicular with 73.23% vesicular ovarian rate, capsular thickening, significantly decreased number of corpora lutea and granulosa cells, and increased theca cells in model group 1(P<0.01). The levels of testosterone(T), luternizing hormone(LH), fasting insulin(FINS) in model group 1 all increased significantly(P<0.05), the level of progesterone(P) decreased significantly in model group 1 (P<0.05), while the level of follicle stimulating hormone(FSH) decreased in model group 1 but had no statistical significance(P>0.05). 70% rats in model group 2 showed estrogen cycle disorders, Compared to control group 2, the weight of the rats and two ovaries were increasing, the ovarian showed multiple follicular with 54.29% vesicular ovarian rate, decreased number of corpora lutea and granulosa cells, and increased theca cells in model group 2(P<0.01), the level of LH, FINS increased significantly in model group 2(P<0.05), while the level of T, P increased in model group 2 with no statistical significance(P>0.05).Conclusion: Base on the Rotterdam polycystic ovarian syndrome diagnostic, from the serum level of hormones and ovarian histology can be obtained that the rat model with polycystic oary syndrome induced by letrozole is better than that induced by HCG combined with INS. Objective: To observe the effect of combination of traditional Chinese medicine and clomiphene on the serum level of sex hormone, the expression of apoptosis factors Bax, Bcl-2 in ovarian antral follicle and leptin receptors in ovarian in PCOS rats, and discuss the mechanism.Methods: The rats with polycystic ovarian syndrome induced by Letrozole were randomly divided into the model group(group M), the Chinese traditional medicine group(group Z), the western medicine group(group X) and the combination of Chinese traditional and western medicine group(group Z+X), each group contained 10 rats. Meanwhile, a group of 10 rats was considered as the control(group N). the treatment group were administered half-dose Letrozole to maintain model. The group M and group N were administered normal saline (NS) at the dose of 1 ml/250 g/d once daily, the group Z was administered Traditional Chinese Medicine (TCM) at the dose of 12.2 g/kg/d, the group X was administered clomiphene citrate (CC) at the dose of 4.5 mg/kg/d, the group Z+X was administered both TCM and CC. the treatment period was two weeks. To weigh the ovarian, to observe follicle change by ovarian histology, the serum level of hormones by RIA, the expression of apoptosis factors Bax, Bcl-2 in ovarian antral follicle by immunohistochemical method and leptin receptor mRNA in ovarian by RT-PCR.Results: Compared to the group N, the level of T, LH, FINS increased significantly (P<0.01)while the level of FSH decreased significantly(P<0.01) in group M, the expression of apoptosis factors Bax and leptin receptor mRNA in ovarian increased while the expression of apoptosis factors Bcl-2 decreased in group M(P<0.05). Compared to the group M, the level of T, FINS decreased significantly in the group Z, X and Z+X(P<0.01), the level of LH decreased in group Z(P<0.05), the level of FSH increased distinctly in group Z, X and Z+X(P<0.05). Compared to the group M, the expression of apoptosis factors Bax and leptin receptor mRNA in ovarian decreased while the expression of apoptosis factors Bcl-2 increased in group Z, X and Z+X(P<0.05), and the expression in the group Z+X was much closer to the group N.Conclusion: The group Z, X and Z+X all can adjust disorder endocrine level of PCOS rats, and promote ovarian development, the effect of group Z+X is the most evident in all groups. TCM combined with CC may improve the inner environment of ovarian development in PCOS model rats by reducing ovarian granulosa cell apoptosis and decreasing the expression of leptin receptor mRNA in the ovary. |
