Aim:Observation and analysis of genes expression ofβ3 adrenergic receptor and uncoupling protein UCP1 in brown adipose tissue between DIO sensitive and resistant ratsMethods:From April 2009 to August 2009, in laboratory of Gastroenterology, department Wuhan Union Hospital.20 SD rats were raised to make DIO rats model, 5 SD rats were given normal diet of ordinary, as the control group. 15 rats were given high-fat, high sugar diet for making DIO sensitive and resistant rat model. Brown adipose tissue was taken from inter-scapular Organization of the rats after Killed by cervical. Tissue was stored at -80℃for RNA extraction. Then total RNA was extracted for RT-PCR, after that cDNA was used for fluorescence quantitative PCR (SYBGREEN method) to detectβ3 adrenergic receptor gene and uncoupling protein UCP1 gene expression. Data of the weight of rats and the genes expression was analyzed by SPSS.Results:9 rats were DIO sensitive; the remaining 6 were DIO resistant. the average weight gain in control group was 75.00±25.00g; the average weight gain in obese group 112.22±23.86g; in resistant group was 38.33±16.33g. There were significantly different compared with the control group. (P value in the table) relative expression ofβ3 adrenergic receptor in brown adipose tissue: the control group was 1.45±0.98; obese group was 0.42±0.36; resistant group 3.20±0.53. ucp1 relative expression of each group: control group 2.13±0.44, obese 1.04±0.36, resistance group 4.12±1.37. There were also significantly different compared with the control group. (see p value in table) .Conclusions:High-fat, high carbohydrate diet can induce obesity SD rats, but there are differences between different individuals. Rats in this test can be divided into obesity-sensitive group and the resistance group. Gens expression ofβ3 adrenergic receptor and uncoupling protein ucp1 were decreased in obese sensitive group and increased in resistant group.β3 adrenergic receptor uncoupling protein ucp1 in brown adipose tissue in rats play the role of resistance in obesity.
