【Objective】To observe the effects of fasudil,a Rho/ROCK signaling pathways inhibitor,on adhesion,migration and the changes in endothelial cytoskeletal reorganization.【Methods】HUVE cells were cultured in vivo.Cultured HUVEC were divided into 5 groups.Fasudil was added to 4 groups in a concentration of 40,70,100,120umol/L,respectively.The effect of Fasudil on proliferative activity of HUVE cells were measured by MTT,calculated the IC50.The inhibition of migration of HUVE cells was evaluated by a transwell chamber.The effect of Fasudil on the changes in endothelial cytoskeletal reorganization were evaluated by phalloidin.The expression of RHOA,PIGF,MMP-9 were measured by Real-time PCR.The protein levels of RHOA,p-MLC(Thrl8/Serl9) were assayed by Western blot.【Results】The IC50 of Fasudil were 867.12±12.6umol/L.The number of cells passed through the Matrigel membrance fell from 253.65±3.21 to 133.32±1.25,(P<0.01).The stress fiber decreased in amount obviously. HUVE cells express RHOA. Expression status of RHOA and adhesion and invasion ability have a positive correlation.【Conclusions】Human umbilical vein endothelial cells have RHOA, PIGF, MMP-9. Fasudil can significantly reduce the invasion and matastatic of HUVE cells in vivo.
