| Object: The research on the chemical composition of Saussurea medusa Maxim is not only have the meaningful guideline to the new drug's quality control,but also provide a scientific basis for further development of Saussurea medusa Maxim. Method: Extract the Saussurea medusa Maxim by supercritical carbon dioxide extraction. The one part of extractant was used to analysis the volatility composition by GC-MS. The others were extracted by petroleum ether and acetic ether. Monomers will be seperated from those two parts using column chromatography of silica gel. After the extacting by supercritical carbon dioxide extraction, Gruffs will be extacted by ethanol refluence extaction for monomers which will be seperated by versamid column chromatography and column chromatography of silica gel.Some spectrums such as IR,EI-MS,1H-NMR,13C-NMR and so on are apply to identify the isolated chemical constituents. Results: The model of Response Surface Methodology was established. 127 volatility compositions which mainly contain fatty acid and ester, alkane,alcohol,aldehyde and aromatic compounds was identified by GC-MS. The Naphthalene,decahydro-2,6-dimethyl,heptacosane,nonacosane,1,6-Dimethylspiro[4.5] decane,Cyclohexene,1-phenyl-and4,4,6a,6b,8a,11,12,14b-Octamethyl-1,4,4a,5,6,6a,6b,7,8,8a,9,10,11,12,12a,14,14a,14b-octadecahydro-2H-picen-3-one were the main volatility composition. On the part of chemical constituents separation,12 compounds were isolated from different polarity parts,10 compounds were identified as follows by spectral analysis: Cerotic acid(2),β-Sitosterol ( 3 ) ,Genkwanin ( 4 ) ,Umbeliferone ( 5 ) ,Scopoletin( 6 ) ,Isoscopoletin ( 7 ) ,Quercetin ( 9 ) , acacetin-7-O-α-L-rhamnopy ranosyl(1→6)-β-D-glucopyranoside(10),Rutin(11), Mannitol(12).The compounds Cerotic acid , Genkwanin, Isoscopoletin, acacetin-7-O-α-L-rhamnopy ranosyl(1→6)-β-D- glucopyranoside and Mannitol were isolated from the plant for the first time. Conclusion: The model of Response Surface Methodology was established and the good effect parts were accumulated for the chemical constituents study. From the volatility composition and the unvolatility composition, the chemical constituents of supercritical carbon dioxide extraction of the Saussurea medusa Maxim was analysised and identified and the chemical constituents of gruffs was studyed preliminaryly. |
