Objective To observe the effect of Electro-acupuncture stimulation on VD rats'Learning-memory and Hippocampus synapse plasticity. We research three kinds of hippocampus area protein : protein kinase C (PKC),synaptophysin(Syp),metabolism glutanic acid acceptor(mGluRs); also includes to research the expression of PKC mRNA .Through those expressions' change ,we want to see the molecular biology mechanism of Electro-acupuncture therapy on VD rats'Learning-memory and Hippocampus synapse plasticity. Then provides certain rationale for the clinical practice Electro-acupuncture treatment.Methods SD rats were randomly divided into Uncontrolled group, Model group and Treatment group.The model group and the treatment group were established Cerebral ischemia model . After that the treatment group were given the certain frequencies acupuncture intervention. Morris water labyrinth were used to test the three groups rats'Learning-memory ability; the immunity histochemistry method detects different groups'Syp,PKC,mGluRs expression; RT-PCR examines the PKC mRNA content; Western blot examines the expression difference of NMDA acceptor .Results Compared with the model group, the treatment group find the platform use a litter time and through platform numbers increased(P<0.05); the expression of Syp and PKC have increased ,but mGluRs have down; the expression of PKC mRNA also have increased; NMDA acceptor have a significant difference between groups.Conclusion Electro-acupuncture therapy could improve Learning-memory ability of VD models. The change expressions of Syp, PKC, mGluRs ,NMDAR may relate to the synaptic plasticity and the change of Long-term potentiation .That give us parts of mechanism in Electro-acupuncture therapy on VD rats'Learning-memory and Hippocampus synapse plasticity in some ways.
