Objective:By comparing the tumor volume, tumor weight, weight of nude mouse and water channel-4 (aquaporin-4 AQP4) level in tumor tissues after usage of different dose of rapamycin on planted subcutaneous glioma in nude mouse, to investigate whether rapamycin affect the growth of planted subcutaneous glioma in nude mouse and the AQP4 level in tumor tissues, whether it is in a dose-dependent manner,and to provide experimental evidence of Rapamycin treatment for glioma.Method:C6 rat glioma cells were cultured (4malignant) to exponential phase (25×107 cells/ml),40 male BALB/c nu/nu mouse (nude mouse homozygote) of 4 to 6 weeks old were planted with C6 cells, each mice was planted subcutaneously at the left and right behind the axillary with 0.2 ml glioma C6 cell suspension (each nude mice was injected with 4~5×107 C6-cells), thus rat xenograft glioma models in nude mouse were establish.10d later,40 nude mouse all bearied the tumor, tumor-bearing nude mouse were randomly divided into four groups, they were given different injection:conventional dose rapamycin group (1.5 mg/kg/d) (n=10), low dose group (0.15 mg/kg/d) (n= 10), high dose group (4.5 mg/kg/d) (n=10), they were given intraperitoneal injection of rapamycin (rapamycin was dissolved in DMSO), in accordance with 0.1ml of rapamycin/one nude mice intraperitoneal injection; each nude mice of control group (n=10)was given the same volume of dimethyl sulfoxide (DMSO) by intraperitoneal injection once a day. During the drug intervention,the general living conditions of nude mouse and volume change of subcutaneous planted tumors were closely observed,after drug intervention of 21d,the 40 experimental nude mouse were sacrificed simultaneously to measure the volume of subcutaneous planted tumors, the tumor weight,inhibitory tumor rate were calculated; Weighing the weight of nude mouse; AQP4 level in subcutaneous planted tumor tissues were detected by immunohistochemistry applicated rabbit anti-rat AQP4 monoclonal antibody.Results:Compared with control group:conventional dose of rapamycin significantly inhibited the growth of subcutaneous planted tumors (p<0.05),the tumor volume, tumor weight of low dose group and high dose group were lower than those of control group,but they were not significant (p>0.05), and conventional dose of rapamycin reduced the AQP4 level in subcutaneous planted tumors (p<0.05),the AQP4 level in subcutaneous planted tumors of low dose group and high dose group were subject to a certain degree of inhibition, but they were not significant (p>0.05),in the experiment,it was also found that the animals in rapamycin conventional dose group tolerated the drug well,their general living conditions were not significantly affected,They were free to eat and the activities; the animals in control group exhibited somewhat weaker activity; the general living conditions of nude mouse in high dose rapamycin greatly affected, Occurred around nude mouse in the drug 8d poor appetite,irritability,skin thin,weight loss and other symptoms, body weight of nude mouse decreased significantly (p<0.05).Conclusion:1, conventional dose of rapamycin can inhibit the growth of planted subcutaneous glioma in nude mouse in a non-dose-dependent and does not significantly affect the general living conditions of nude mouse; low-dose and high dose of rapamycin could not inhibit the growth of planted subcutaneous glioma in nude mouse;nude mouse can not tolerate high dose of rapamycin.2, conventional dose of rapamycin can inhibit the expression of AQP4 of planted subcutaneous glioma in nude mouse and in a non-dose-dependent manner.
