Objective; To evaluate expression of netrin-1 protein in retina of mice model of oxygen induced retinopathy (OIR).Methods; 16 C57BL/6J newborn mice were involved in this study. Mice randomly divided into normal control group and oxygen induced retinopathy (OIR) group. (n=8). In OIR group, mice were exposed to 75% oxygen to induce oxygen induced retinopathy (OIR) model. Then,two mice from each group were selected to be made retina flatmount to observe the retinal neovascularization for identifying Oxygen induced retinopathy model.The remained mice were used to check the expression of Netrin-1 protein in the retina through immunohistochemistry.Results; (1)The analyzing of retinal fluorescence imaging; Normal mice retina showed retinal blood vessels distributed regularly. In hyperoxia-induced group, there were a large area of retina without perfusion surrounding the optic disc area and a large amount of retinal neovascularization with fluorescein leakage. (2) Immunohistochemistry Results; There was no expression of netrin-1 protein in the normal group, in contrast, expression of netrin-1 at the superficial retina in the oxygen induced group.Conclusion; The expression of Netrin-1 at the stage of retinal neovascularization under OIR conditions suggested that Netrin-1 may be involved in the formation of retinal neovascularization.
