Objective To study the expression of Ror2 and Wnt5a protein in human osteosarcoma and the correlation between them. So can investigate their significance in osteosarcoma`s occurrence and development.Methods 55 cases of human osteosarcoma specimen were selected in the study, compared with 15 cases of osteochondroma. We detected the expression of Ror2 and Wnt5a of osteosarcoma by S-P immunohistchemistry method and analysis the expression relationship with soft ware of SPSS 17.0.Results (1)The positive expressive rate of Ror2 in osteosarcoma was 70.91%(39/55),while in osteochondroma it was 20.00%(3/15), the rate of osteosarcoma is higher than that of osteochondroma.There were statistical significant difference betreen the two groups (χ2=12.73, P<0.05). (2)The positive expressive rate of Wnt5a in osteosarcoma was 74.55% (41/55),while in osteochondroma it was 13.33%(2/15),the rate of osteosarcoma is higher than that of osteochondroma. There were statistical significant difference betreen the two groups (χ2=18.64, P<0.05). (3)The positive expressive rate of Ror2 in stageⅡandⅢwere remarkably higher than that of stageⅠ, there were statistical significant difference between the two groups (P<0.05). While the positive rate had no significant difference between stageⅡand stageⅢ(χ2=2.85, P>0.05). The positive expressive rate of Ror2 in osteosarcoma with metastasis was 100.0% (13/13),while in osteosarcoma without metastasis it was 61.90%(26/42), there were statistical significant difference between the two groups (χ2=5.26, P<0.05). (4)The positive expressive rate of Wnt5a in stageⅡandⅢwere remarkably higher than that of stageⅠ, there were statistical significant difference between the two groups(P<0.05).While the positive rate had no significant difference between stageⅡand stageⅢ(χ2=1.35, P>0.05).The positive expressive rate of Wnt5a in osteosarcoma with metastasis was 100.0%(13/13),while in osteosarcoma without metastasis it was 66.7%(28/42), there were statistical significant difference between the two groups (χ2=4.19, P<0.05). (5)The positive expressive rate of Ror2 and Wnt5a in osteosarcoma had no significant among different sex, age and patho-logical types (P>0.05). (6)The was positive correlation between the expression Ror2 and the expression of Wnt5a in osteosarcoma (r=0.831, P<0.01).Conclusions(1)The protein of Ror2 and Wnt5a was in highly level expressed in osteosarcoma and lower expressed in osteochondroma.(2) In the Ennecking surgical staging of osteosarcoma, the positive expressive rate of Ror2 and Wnt5a in stageⅡandⅢwere remarkably higher than that of stageⅠ, the expression of Ror2 and Wnt5a in osteosarcoma's occurrence and development play an important roel, and correlation with the degree of malignancy osteosarcoma.(3)Ror2, Wnt5a in the metastasis of osteosarcoma tissues is far higher than those without metastasis of osteosarcoma, the evidence indicate that Ror2 and Wnt5a may correlation with osteosarcoma invasion and metastasis.(4) In osteosarcoma tissue, Wnt5a is positively correlated with the expression of Ror2, the evidence indicate that Wnt5a and Ror2 protein in invasion and metastasis osteosarcoma is synergy process.(5) The positive expressive rate of Ror2 and Wnt5a in osteosarcoma had no significant among different sex, age and patho-logical types.
