Objective:(1) To test the expression of Kruppel-like Factor 4 in patients with CML (chronic myeloid leukemia) and to discuss the role played in pathogenesis of CML. (2) To test the expression of klf4 mRNA and KLF4 Protein in leukemic cell line K562 and to discuss their role in pathogeny of CML.Method:(1) The relative expression of klf4 mRNA in 32 patients with preliminary diagnosed CML, who is in the stage of chronic phase with fusion gene of bcr/abl, and 18 healthy controls were tested with Rverse Transcription-polymerase chain reaction. (2) Expression of klf4 mRNA and KLF4 Protein in leukemic cell line K562 were tested with Fluorescence Quantitative polymerase chain reaction and Wetern blot technology.Result:(1) Klf4 mRNA is expressed in both patients with CML-CP and healthy controls.The relative expression of klf4 mRNA (0.095±0.031) in 32 patients with CML as significance difference compared with healthy controls (0.220±0.046) (p<0.05). (2) Both klf4 mRNA and KLF4 protein are expressed in leukemic cell line K562.Compared with control group(0.02±0.01), the expression of klf4 mRNA in K562 cell was significance difference(p<0.05). The expression of KLF4 protein in K562 cells is much lower than that in human peripheral monocyte/macrophage.Conclusion:(1) Compared with healthy controls, the expression of klf4 gene in patients of preliminary diagnosed CML-CP with fusion gene of bcr/abl is lower. (2) There is the low expression of klf4 mRNA in leukemic cell line K562.lt is obvious that the expression of KLF4 protein in human peripheral monocyte/ acrophage is higher than that in K562 cells by Western blot technology. (3) It suggests that KLF4 maybe partially involved in the mechanism in leukemogenesis.
