| Objective: Through the establishment of rat tongue mucosa carcinogenesis model and application of Qi-lan particles to intervene to observe the tumor suppressor role in blocking cancer while using immunohistochemical SABC method of proliferating cell nuclear antigen (PCNA), epidermal growth factor receptor (EGFR), P27 protein, changes in the expression. Qi-Lan particles obstructing carcinoma of the possible mechanism of tumor suppressor.Methods: Animal model selected 6~7-week-old SD rats were 150, being randomly divided into five groups (30 rats / group), A group for cancer model group, B group for low-dose intervention group, C group, the dose of the intervention group, D group,high-dose group intervention group, E group, normal control group. Drinking Water act administration,the rats were given 0.002% 4NQO and 0.002% 4NQO plus low, medium and high doses of three kinds of Qi-lan particles to establish cancer rat model and drug intervention model. At 9 weeks, 18 weeks, 27 weeks, 36 weeks, animals were sacrificed in batches (7 rats/week), taking the organization to do the tongue biopsy, using immunohistochemical SABC assay PCNA, EGFR, p27 protein expression, and through statistical software, the experimental results were analyzed statistically.Results:1. Qi-Lan particles with a centralizer and the obvious strengths of the effect of tumor suppressor cancer resistance: A group of rats relative to B, C, D rats fatigue lethargy, fur color yellow, rough dull, body weight gradually reduce to 27,36 week, A, B, C, D groups were statistically significant differences in cancer levels, A group of rats, the overall cancer rate in the B, C, D group is high (p <0.05), in which the cancer group the lowest level of C ( p <0.01).2. At all levels, from normal oral mucosa→dysplasia→the process of squamous cell carcinoma lesions, PCNA, EGFR protein expression increased progressively (p<0.05), p27 protein expression showed decreasing trend (p<0.05).3. A tongue tissue dysplasia group PCNA and EGFR protein expression in the intervention group was higher than that of Chinese medicine in each group (P <0.05), A group of p27 protein expression in the intervention group were significantly lower than that of Chinese medicine in each group (p<0.05); tongue tissue A group of PCNA protein and p27 protein expression with traditional Chinese medicine interventions had no significant statistical difference between groups (p> 0.05), A group of EGFR protein expression in the intervention group were significantly higher than that of Chinese medicine groups (p <0.05).4. In abnormal proliferation of tissue PCNA protein expression and p27 protein expression was negatively correlated (r=- 0.786, p <0.001), PCNA protein expression and EGFR protein expression was positively correlated (r = 0.588, p <0.001); in the cancerous tissue in the PCNA P27 protein expression and protein expression was negatively correlated (r =- 0.502, p <0.05), EGFR protein and cell proliferative activity is not related (r = 0.252, p> 0.05).Conclusion:1, Qi-lan can reinforce the efficacy of particles with a centralizer.2, Qi-lan particles can significantly reduce the tongue mucosa dysplasia and cancer incidence, delaying the process of tongue cancer, on experimental carcinogenesis of oral mucosa has obvious resistance cancer, tumor suppressor role.3, PCNA, EGFR and p27 expression and the occurrence of oral squamous cell carcinoma development can be used as assessment of the status of oral squamous cell carcinoma cell proliferation marker.4, phase dysplasia, EGFR can promote cell proliferation, p27 could inhibit cell proliferation, regulation of mutual restraint between the two involved in tumor formation; malignant tumor stage, p27 on the progress of cancer is still limiting.5, Qi-lan particles cancer resistance mechanisms may, in the abnormal proliferation phase by down-regulating the expression of PCNA and EGFR protein in the same time, increase the expression of p27 protein, thereby inhibiting abnormal proliferation of epithelial cells, play a role in blocking cancer, and in cancer the organization by down-regulating the expression of EGFR play a role in inhibiting cancer... |
