Objective To investigate the expression of Phosphatase and tension homology deleted from chromosome 10(PTEN) mRNA,Survivin mRNA and vascular endothelial growth factor(VEGF) mRNA in tissues of bladder transitional cell carcinoma (BTCC) of different pathological grades,clinical stages,primary and recurrence, and their clinical significance.Methods The SYBR Green I real time quantitative RT-PCR was used to detect the expression of PTEN mRNA,Survivin mRNA and VEGF mRNA in 51 cases of transitional cell carcinoma of bladder tissues and 12 cases of normal bladder tissues.All of data were treated statistically.Results The expression of PTEN mRNA in normal bladder tissues was higher than that in BTCC tissues, while the expression of Survivin mRNA and VEGF mRNA in normal tissues was lower than that in BTCC tissues. The comparison of all of them between normal bladder tissues and BTCC tissues has statistical significance respectively(P <0.05). The expression of PTEN mRNA in low grade BTCC tissues was higher than that in high grade BTCC tissues(P <0.05), while the expression of Survivin mRNA and VEGF mRNA in low grade BTCC tissues was lower than that in high grade BTCC tissues respectively(P <0.05). The expression of PTEN mRNA in superficial BTCC tissues was higher than that in invasive BTCC tissues(P <0.05), while the expression of Survivin mRNA and VEGF mRNA in superficial BTCC tissues was lower than that in invasive BTCC tissues respectively(P <0.05). The expression of PTEN mRNA in primary BTCC tissues was higher than that in recurrence BTCC tissues(P <0.05), but the expression of Survivin mRNA and VEGF mRNA in primary BTCC tissues was lower than that in recurrence BTCC tissues respectively(P <0.05). There were negative correlations between the expression of PTEN mRNA and Survivin mRNA(r=-0.819,P <0.05) and PTEN mRNA and Survivin mRNA (r=-0.845,P <0.05)in BTCC tissues, and there was a positive correlation between the expression of Survivin mRNA and VEGF mRNA(r=0.889,P <0.05).Conclusions The low expression of PTEN mRNA and high expression of Survivin mRNA and VEGF mRNA in BTCC tissues may play important roles in the pathogenesis and development of BTCC, and all of them can be considered as important indexes to judge the biological behaviors of BTCC. Combined detection of PTEN mRNA,Survivin mRNA and VEGF mRNA would be helpful to evaluate the condition and prognosis of patients with BTCC.
