Objective To investigate the expression of eIF4E(eukaryotic translation initiation factor 4E) and PTEN(Phosphatase and tensin homology deleted on chromosome ten) in human lung adenocarcinoma tissue, to seak their relationship in clinicopathological features; to explore the relationship between eIF4E and PTEN expression and clinicopathological features in human lung adenocarcinoma; and to find out the relationship between eIF4E and PTEN in human lung adenocarcinoma. Method The expression of PTENmRNA and eIF4EmRNA was examined by reverse transcriptase poly merase chain reaction(RT-PCR) analysis in 40 cases of human lung adenocarcinoma and 20 cases of normal lung tissues and its correlations of clinicopathological features were analyzed. The expression of PTEN and eIF4E in 40 cases of lung adenocarcinoma and 20 cases of normal lung tissues were detected by immunohistochemistry test (S-P method) and its correlations of clinicopathologic features were analyzed. Results 1.The loss of PTEN expression in lung adenocarcinoma tissues and normal lung tissues was 60.0%(24/40),15%(3/20), respectively. Loss of PTEN expression in lung adenocarcinoma was significantly higher than that in normal lung tissue (P<0.05). The loss of PTEN expression in lung adenocarcinoma was related to degree of cell differentiation tumor and lymph node metastsis (P<0.05), but not to age, sex, TNM stages, (P>0.05). The loss of eIF4E expression in lung adenocarcinoma tissues and normal lung tissues was0%(0/40),100%(20/20), respectively. Loss of eIF4E expression in normal lung tissue was significantly higher than that in lung adenocarcinoma (P<0.05). The expression of eIF4E in lung adeno- carcinoma was related to degree of cell differentiation tumor and lymph node metastsis (P<0.05), but not to age, sex, TNM stages (P>0.05).2. Immunohistochemistry showed that the positive rate of PTEN in lung adenocarcinoma tissues and normal lung tissues was 45.0%(18/40),90.0%(2/20), respectively. The positive rate of eIF4E in lung adenocarcinoma tissues and normal lung tissues was 60.0%(24/40),0%(0/20), respectively. With the degree of cell differentiation tumor and lymph node metastsis being higher, the lower expression level of PTEN showed(p<0.05), while the expression of eIF4E increased (p<0.05) (3)There was negative correlation between PTEN protein and eIF4E protein expression in lung adenocarcinoma tissues ((r=-0.427, p<0.05). Conclusion 1. The abnormal expression of suppressor gene PTEN and oncogene eIF4E Participate the malignant conversion process of the lung adenocarcinoma and correlation with tumor histological poor differentiation and lymph node metastasis'; the expression levels of the PTEN and eIF4E protein can be used to evaluate the pathologic biology behavior of lung adenocarcinoma.2. The loss of PTEN may up-regulated the expression of elF4E though PI3K/mTOR/4E-BPs/eIF4E signal transduction pathway. It may play an important role in genesis and growth of lung adenocarcinoma.
