| Objective To observe the effects of butylphthalide on the expression of NeuN in the CA1 region of hippocampus and IL-1 in the junction of cortex and CA1 region of hippocampus in model rats with Alzheimer disease (AD), and explore the potential mechanism of butylphthalide on AD.Methods Sixty healthy male Sprague-Dawley (SD) rats were selected and then randomly divided into model group, butylphthalide group, control group. The AD model was established by microinjection of beta-amyloid peptide 1-42 (Aβ1-42) into the left hippocampus stereotacticly. The learning and memory ability were evaluated by Y-type electric maze before and sixty days after operation. The morphology of neurons in the CA1 region of hippocampus was observed by HE staining. The expression levels of NeuN and IL-1 were measured by immunohistochemistry (IH)Results1. the learning and memory abilities of AD model ratsIn the test of Y-type electric maze, comparison among groups showed no significance (F=0.014, P=0.986) before operation. Sixty days after operation, significance was showed among groups (F=24.337, P<0.001). Comparison between the model group and the control group showed significance (P<0.001), the model group's time to evade stimulate is more. Comparison between the butylphthalide group and the model group showed significance (P<0.001), the butylphthalide group's time to evade stimulate is less. Comparison between before the operation and after the operation of the model group and the butylphthalide group showed significance (P<0.001), which of the control group showed no significance (P=0.248>0.05)2. Expression of NeuNThere was significant difference among groups (F=289.156, P<0.001). Compared with the control group, the model group showed significant difference (P<0.001), the expression of of NeuN of the two groups are different.Compared with the model group, the butylphthalide group showed significance (P<0.001)3. Expression of IL-1There was significant difference among groups (F=584.205, P<0.001). Compared with the control group, the AD group showed significant difference (P<0.001), the expression of of IL-1 of the two groups are different. Compared with the AD group, the butylphthalide group showed significance (P<0.001)Conclusions1. The expression of NeuN decreased and the expression of IL-1 increased in AD rats.2. Butylphthalide has an effect of improving learning and memory ability of AD model rats, which is possibly due to the reduce of the expressions of IL-1 and the protection of the nerve tissue. |
