Objective:To explore the function of Caveolin-1 and Cyclin D1 protein in the development and metastasis of gestational trophoblastic tumor by analyzing the expression of them in normal early pregnancy villi and gestational trophoblastic diseases and reaserching the relationship of them with clinical and pathological factors of gestational trophoblastic tumors.Methods:The expression of Caveolin-1 and Cyclin D1 protein were detected by immunohistochemistry(S-P) in normal pregnancy villi (20 cases),hydatidiform moles(35 cases),invasive hydatidiform moles(18 cases) and chorionic carcinomas (12cases).Results:1.The positive rate of Caveolin-1 in normal pregnancy villi,hydatidiform moles,invasive hydatidiform moles and chorionic carcinomas were respectively 30.0% (6/20),40.0%(14/35),72.2%(13/18),83.3%(10/12). The difference between normal pregnancy villi group and invasive hydatidiform moles group, between normal pregnancy villi group and chorionic carcinoma group, between hydatidiform moles group and invasive hydatidiform moles group, and between hydatidiform moles group and chorionic carcinoma group were statistically significant (P=0.022, P=0.009, P=0.026, P=0.010).The expression of Caveolin-1 was correlated to clinical stage, while there was no close relation to ages.2. The positive rate of Cyclin D1 in normal pregnancy villi, hydatidiform moles, invasive hydatidiform moles and chorionic carcinomas were respectively5% (1/20),22.9%(8/35),66.7%(12/18),75.0%(9/12). The difference between normal pregnancy villis group and invasive hydatidiform moles group, between normal pregnancy villis group and chorionic carcinoma group, between hydatidiform moles group and invasive hydatidiform moles group, and between hydatidiform moles group and chorionic carcinoma group were statistically significant (P=0.000,P=0.000,P=0.002,P=0.001).The expression of Cyclin D1 was no close relation to clinical stage or ages.3. There was a positive relationship between the protein expression of Caveolin-1 and Cyclin D1 in GTN(r=0.291, P<0.01).Conclusion:1.The expression of Caveolin-1 and Cyclin D1 in gestational trophoblastic tumor are higher than in normal pregnancy villi and hydatidiform moles, which indicates that Caveolin-1 and Cyclin D1 may play a role in the progress of malignant transformation of trophoblastic cells.2.The expression of Caveolin-1 is possibly related to clinical stage, and is likely to promote the invasion and transformation of gestational trophoblastic tumor.3.The expression of Caveolin-1 and Cyclin D1 may work in coordination to promote the development of GTN.
