| Objective and meaning: Asthma is an inflammatory disorder of the lung that affects people of all ages. The morbidity and mortality associated with asthma have increased sharply all around the world over the last 40 years. Because of high morbidity,mortality and financial burden, asthma has already been a problem which all people of the world need to face.Asthma is a chronic inflammatory disorder of the airways in which many cells and cellular elements play a role and chronic inflammation of airway, reversible bronchial obstruction, airway hyperresponsiveness are its features. The abnormal accumulation and activation of Eos is considered a main reason of asthma. Many cytokines and chemokines, especially IL-5, play an important role in differentiation, maturation, survival and migrating to the circulatory system of Eos.IL-5 can increase the number of Eosinophil (Eos) of airway through IL-5 dependent and independent mechanism. IL-5 independence mechanism which mainly stimulate the bronchial airway epithelial by IL-13. The two different mechanisms have an integrated role in allergic disease and do not exclude each other. So far, the mechanism of AHR is unknown.The effects of airway inflammation, such as Eosinophil, Macrophages, lymphocytes and neutrophils, and airway hyper- responsiveness(AHR)were observed by using anti-IL-5 antibody, anti-IL-13 antibody and anti-IL-5 antibody combined anti-IL-13 antibody in a rat asthmatic model, which was established a foundation for clinical application.Methods:â‘ 50 female Balb /c rats were divided randomly into 5 groups: the normal control group , the asthmatic model group, the anti-IL-5 antibody group, the anti-IL-13 antibody group and anti-IL-5 antibody combined anti-IL-13 antibody group.â‘¡A rat asthmatic model was established.â‘¢The Eosinophil, neutrophil, lymphocyte count in bronchoalveolar lavage fluid (BALF) were made with Wright's staining.â‘£The change of the airway resistance was analyzed by means of animal lungs function analysis system.Results:â‘ Anti-IL-13 antibody group relieve the symptom of asthmatic model gentlely compared with asthmatic model group. Onset of asthmatic symptom of anti-IL-5 antibody group, such as irritability and breathless is alleviate slightly compared with asthmatic model group. Onset of asthmatic symptom in anti-IL-5 antibody combined anti-IL-13 antibody group can be relieved compared with the asthmatic model group and other treatment groups. The performance of normal group was good as usual.â‘¡Levels of total cellular, Eosinophil, neutrophil, lymphocyte in BALF of asthmatic group were significantly higher than the normal group (P<0.01). Levels of total cellular and inflammatory cells of anti-IL-5 antibody group were decrease slightly compared with the asthmatic group. (P<0.05). There was no difference between asthmatic group and anti-IL-13 antibody group (P<0.05). Levels of total cellular, Eosinophil, neutrophil, lymphocyte in BALF were significantly declined in anti-IL-5 antibody combined anti-IL-13 antibody group than the other treatment groups (P<0.05);â‘¢Airway resistance of asthmatic group stimulated by Methacholine was higher than normal group and there was no difference between asthmatic group and anti-IL-13 antibody group(P>0.05);The level of Airway resistance can be markedly reduced by anti-IL-5 antibody combined anti-IL-13 antibody, which has statistically significant difference compared with other treatment groups(P<0.05).Conclusion:â‘ Compared with the asthmatic group, anti-IL-5 antibody combined anti-IL-13 antibody can decrease Eos, neutrophil and lymphocyte infiltrationâ‘¡Anti-IL-5 antibody combined anti-IL-13 antibody can alleviate AHR and improve pulmonary function in rat asthmatic model, which may be one of effective way to treat asthmatic disease. |
