| Diospyros kaki (Diospyros kaki Thunb.) is an economic species in hunan province. Recently, several sweet Diospyros kaki cultivated varieties are introducted successfully in hunan province. Diospyros kaki is very useful all of it. Diospyros kaki leaves is an important Chinese medicine resources. It has higher nutritional value and medical effect. Now research proved that Flavonoids is main effective component in Diospyros kaki leaves, In this paper, by surveying and sampling the Diospyros kaki resources in hunan province to analyze and compare different varieties'fruit quantity; by using HPLC analysis method and spectrophotometry method to survey research the Flavonoids chemical resources of Diospyros kaki leaves in hunan province; by using HSCCC and preparative HPLC to separate and prepare Flavonoids from Diospyros kaki leaves' extraction and identify them. The main research results are as followed:1 Survey research of Diospyros kaki resources in hunan provinceBy analyzing fruit quantity and Flavonoids and Carotenoids contents from leaves of different Diospyros kaki varieties and production area in hunan province to select the varieties of higher comprehensive using value. The results are as followed:Xinjin20 and Sandaijin have better fruits' quantity, there are all introducted varieties in hunan province. And their leaves also have higher content of effective component. They are the good varieties to fit to extend and plant in hunan province.2 Study on flavonoids chemical resources of Diospyros kaki leaves by HPLC in Hunan provinceBy setting up the HPLC analysis method of Flavonoids from Diospyros kaki leaves and using spectrophotometry to analyse and compare the Flavonoids contents from Diospyros kaki leaves of different picking time and varieties in Hunan province. The HPLC analysis method in this paper is as followed:C18column(φ4.6mm×150nm, 5μm);column temperature is 30℃; detection wavelength is 360nm; the flow rate is 1mL/min;inject volume is 20pL; the mobile phase is acetonitrile-0.1% phosphoric acid water,0-30 min 15% acetonitrile,30-60min 20% acetonitrile,60-70min 30% acetonitrile,70-85min 30% acetonitrile,85-90min 80% acetonitrile.The comparing results are as followed:The reasonable picking time of Diospyros kaki leaves is the last ten days of March to the first ten days of April and the the last ten days of September to the first ten days of October; Flavonoids contents from different varieties in the same area and the same variety in different areas have difference in Hunan province.3 Separate and prepare Flavonoids from Diospyros kaki leaves and identify their structuresBy using HSCCC and preparative HPLC to separate and prepare Flavonoids from Diospyros kaki leaves' extraction; And, using HPLC method to detect the prepared fractions and identify their structures by color reaction and UV spectrum and MS and NMR. The results are as followed:All of the four prepared fractions are Flavonoids, their purity is respectively 98.3%,92.7%,99.1%,91.5%; Their structures maybe is quercetin-3-β-D-galactoside,quercetin-3-β-D-glucoside,kaemoferol-3-β-D-galactoside,kaemoferol-3-β-D-glucoside. |
