| Object:By detecting the expression of PTEN,Survivin and COX-2 proteins in the tissue of non-small cell lung cancer(NSCLC) and non-lung cancer lung tissues. We studied the effects of PTEN,Survivin and COX-2 proteins and the correlation among PTEN,Survivin and COX-2 proteins on the development of NSCLC.And study the relationship between the three protein gene from the molecular mechanism of the anti-apoptosis. Methods:The expression of PTEN, Survivin and COX-2 proteins was detected in 60 cases of NSCLC and 30 cases of non-lung cancer lung tissues by immunohistochemical method.Results:(1) The rate of positive expression of PTEN proteins was higher in the tissues non-lung cancer lung than those in tissues of NSCLC, The rate of positive expression of Survivin and COX-2 proteins was higher in the tissues of NSCLC than those in non-lung cancer lung tissues. (2) The degree of expression of PTEN protein in NSCLC was relative to the clinical stages and metastatic lymphonode, but was not relative to the age, sexuality, the history of smoking, the pathological types, histologic classes and the size of tumor. The later clinical stage and the metastatic lymphonode, the lower rate was the positive expression of PTEN. (3) The degree of expression of Survivin protein in NSCLC was relative to clinical stages and metastatic lymphonode, but was not relative to the age, sexuality, the history of smoking, the pathological types, histologic classes and the size of tumor. The later clinical stage and the metastatic lymphonode, the higher rate was the positive expression of Survivin. (4) The degree of expression of COX-2 protein in NSCLC was relative to the pathological types, histologic classes, the size of tumor, clinical stages and metastatic lymphonode, but was not relative to the age, sexuality and the history of smoking. The rate of expression of COX-2 protein in adenocarcinoma was higher than in squamouscancer; The worse histologic classes, the bigger tumor, the later clinical stage and the metastatic lymphonode, the higher rate was the positive expression of COX-2 protein.(5)The level of expression of PTEN,Survivin and COX-2 proteins was closely related to each other in the carcinoma of NSCLC, the coefficients of correlation were-0.367,-0.299and 0.319 p<0.05.Conclusions:The experimental results were suggested that PTEN,Survivin and COX-2 proteins play important roles in oncogenesis and progression of NSCLC. The lower positive expression of PTEN proteins and the higher positive expression of Survivin and COX-2 proteins was indication for the tumor growing quickly and the disease stage being later and lymphonode having metastasis.The expression of PTEN,Survivin and COX-2 proteins in NSCLC was related to each other. PTEN,Survivin and COX-2 had together taken part in the oncogenesis, development, invasion of carcinoma and lymphonode metastasis in NSCLC.Detection the level of PTEN,Survivin and COX-2 proteins expression might be useful to judge the stage of disease and lymphonode metastasis of NSCLC, and to predict the prognosis of patients with NSCLC. Also, there results have provided a fundamental theory for the prevention and treatment of NSCLC. |
