Studies On Effect Of Reliving Cough And Reducing Sputum Of Pinellia Rhizoma Prepared By Alum And The Quality Standards

This project came from the the first subdivision on 2010 edition "Chinese pharmacopeia" standard research task(132), and was supported partly by national natural science foundation (No.30772785) and "Eleven-Five" national projects of technological supporting (No.2006BAI09B06-10).Pinellia Tuber is the dry tuber of Pinellia ternata (Thunb) Breit. Because of its toxicity, it is mostly processed before it is ready for clinical use. Pinellia Rhizoma prepared by alum through duplicating method is one of the processed products of Pinellia Rhizoma, and has the effects of relieving cough and reducing sputum. Pinellia Rhizoma prepared by alum is classified under the category of Pinellia Rhizoma in Chinese Pharmacopoeia (2005), and it has no objective detection index except for simple character description in this Pharmacopoeia, so its quality couldn't be controlled effectively. On the basis of massive literature research, systematic studies on the processing technology and quality standards of Pinellia Rhizoma prepared by alum were made in this paper. The main research contents and work are as follows: 1 The effect of relieving cough and reducing sputum of Pinellia Rhizoma prepared by alumThe experiment of relieving cough was by the method of cough induced by strong ammonia solution and stimulation of citric acid. Observing the effect of reducing sputum by the method of pushing pheonl red in trachea. The experiment of anti-inflammatory was made by ear edema tests, subcutaneous filter paper and abdominal cavity capillary permeability in mice caused by acetic acid. It was proved that Pinellia Rhizoma Prepared by alum could obviously prolong the latent period and decrease the times of coughing, and notablely increase the capacity of pushing pheonl red in trachea. Pinellia Rhizoma Prepared by alum relieved the mouse ear swelling, reduced capillary permeation and granuloma. Pinellia Rhizoma Prepared by alum has the effect of relieving cough and reducing sputum and anti-inflammatory. There is the same effects between Pinellia Rhizoma Prepared by alum and Pinellia Rhizoma. Pinellia Rhizoma Prepared by alum has stronger effects than Pinellia Rhizoma on effect of anti-inflammatory.2 Components analysis and determination of total free organic acid of Rhizoma Pinelliae Prepared by alum.The total free organic acid of Pinellia Rhizoma prepared by alum was confirmed to have the extinctive activiry of inhibiting coughs. In this paper, potential residual titration was used to determinate the total free organic acids. The total free organic acids were analyzed by GC method. Five kinds of total free organic acids were detected in both Pinellia Rhizoma and Pinellia Rhizoma prepared by alum. The determination of the total free organic acid in Rhizoma Pinelliae Prepared by alum were higher than Rhizoma Pinelliae.3 Qualities of phizoma pinelliae prepared by alum were built upIn this section, the 10 samples of Pinellia Rhizoma prepared by alum from different companies were studied. The pharmacognostical methods and TLC were applied. Moisture, total ash, extract, alum residual amount and total free organic acid of Rhizoma Pinelliae Prepared by alum were determined. The pharmacognostical methods and TLC were applied. The morphological and histological characters of 10 samples from different companies were similar. The suggested quality standard of Rhizoma Pinelliae Preparate is:moisture, not more than 13.0%; total ash, not more than 3.5%; water lsoluble extract, not less than 7.0%; alum residual amount, not more than 8.0%; total free oranic acid, not less than 0.30%. Standard was listed in the first subdivision of 2010 edition "Chinese pharmocopeia".4 Principium stability researchThree batches of Pinellia Rhizoma prepared by alum were conserved in condition of temperature 40℃±2℃and relative moisture 75%±5% for 6 months, observing the quality in 0, 1,2,3,6 month. Make the same three batches of pinelliae ternate by alum in natural condition without light in 12 months and the quality according to quality standard were reviewed when the 0, 3,6,9,12 month. All of them were accorded with quality standard. The result of research indicated that Pinellia Rhizoma prepared by alum could be conserved about 24months.
