Objective:The previous research suggested that Danshensu (DLA) could inhibit metastasis of melanoma. The result of molecular docking suggested DLA might have effect on adhension and invasion of melanoma. Cell adhension and invasion played an important part in melanoma hematogenous dissemination. In order to make further development on DLA anti-tumor mechanism, a series of research were made on melanoma adhension and invasion.Methods:The research of DLA included three chapters, of which previous two mainly studied cell adhension. This section studied whether DLA could affect adhension biologic behavior in melanoma hematogenous dissemination. Adhension behavior included adhension between melanoma and matrigel, ability of cell aggregation and adhension between melanoma and platelet. Based on these researches, relative adhension molecule expression can be learned. The third chapter mainly studied whether DLA could affect invasion biologic behavior which contained degradation of matrigel and cell motion. Adhension:Technology of staining was used to study the adhension between melanoma and matrigel. Cell aggregation assay was used to study melanoma aggregation ability. Flow-cytometer and fluorescence labelling platelet was used to assay adhension between platelet and melanoma. Real-time PCR was used to assay the expression of MCAM,β3 integrin, E-cadherin which reacted in melanoma adhension. Western blot was carried to assay expression of MCAM. ELISA was used to assay sICAM-1.Invasion: Trans-well was used to study melanoma invasion; scratch method was carried to assay cell motion; Real-time PCR was used to assay expression of MMP2, MMP9 and inhibit factor TIMP2 and RECK, Western blot was also used to assay MMP2 and TIMP2.Conclusion:DLA could inhibit melanoma cell aggregation and adhension between melanoma and matrigel, but have no effect on platelet aggregation and adhension between melanoma and platelet. DLA inhibit melanoma adhension molecule expression of MCAM based on mRNA and protein level. Besides, DLA could inhibit A375 releasing of sICAM. The result also suggested DLA inhibit melanoma invasion, and inhibit cell motion and expression of enzyme in invasion process.All these datas suggested DLA inhibit melanoma hematogenous dissemination potentially, and DLA mainly affected melanoma adhension and invasion. This research provided referenced value on anti-tumor metastasis of traditional Chinese medicine DANSHEN so that we can make further development and utilization.
