| Objectives:To observe the interrelation of Content variation of urotensinⅡ(UⅡ) and brain natriuretic peptide(BNP)of rat in different Cardiac function and TCM, and to explore the clinical meaning. Methods:40 male Wistar rats, weight(200±20)g, were randomly divided into chronic heart failure (CHF) model group,30rats; physiologic saline (NS)(normal control group)model group,10 rats.1.CHF model established: from 30 healthy male Wistar rats, weight (200±20) g, The ISO—induced group(ISO group, n=30)received two subcutaneous injections of 170 mg ISO (24 hour intervals)per kilogram of body; 2. the normal, control group(NS group, n=10)received two subcutaneous injections of 0.25 ml normal saline.3. Model identification:(1) Conventional breeding in a same room, to observe the rat diet, fur, feces, activity, cyanosis, edema and the general traits, weighed Weekly, and in accordance with the performance of rats with symptoms and activities, divided rats into type of xin qi failure and type of xin yang failure, Record the time of rat death and mortality.(2) Cardiac function testing:Measurement of left ventricular systolic pressure (LVSP) left ventricular end-diastolic pressure (LVEDP), Maximal rising rate of left ventricular systolic pressure(+LVdp/dt) and Maximum descent rate of of left ventricular diastolic pressure(—LVdp/dt) by Carotid artery.(3)After cardiac function, Removal of rat heart, liver and lungs, calculated separately heart, liver, lung mass index. (4) Biopsy pathology testing:Left ventricular myocardium tissue conventional paraffin-embedded sections, HE staining, microscopy.4. Determination of concentrations of plasma urotensinⅡ(UⅡ) by enzyme-linked immunosorbent assay.5.Determination of plasma brain natriuretic peptide(BNP) by enzyme-linked immunosorbent assay.6.Data Analysis: all of the measurement data are expressed with x±s, and analyzed with SPSS 13.0, test of Homogeneity of variance, One-Way ANOVA, F-test of analysis of variance was used to compare the mean of Multi-groups and X2 Test is applied in the analysis of enumeration data, Correlation analysis using linear correlation analysis, p>0.05 for the difference Was no significance statistically, p<0.05 for the difference Was significance statistically. Result:1.the general state of rats and death rate:compared with NS rats, most of CHF model group rats:dispirited,fatigue,dry fur,relative reduction of activity and eating,temperature drop,Shy and crowding together,shrinking the shoulder extrados dorsal,foot edema,dark purple tongue,and so on. Mortality rate was 20%, and no death of the NS group. the rate of groups was p<0.05,NS rats were no significant change generally.2. Observation of pathology of rat myocardial tissue (HE staining):NS Group:Myocardial fibers were arranged regularly, normal cells gap, no destruction of cardiac fibers; CHF group:myocardial tissue was damaged, Myocardial fiber is disorder, One part of the fracture, hyperemia and infiltrated inflammatory cells can be seen between the myocardial cell, part of myofibrils dissolved and vanished; 3.Plasma urotensinⅡin rats:compare with the NS group, the plasma urotensinⅡin rats of CHF group were significantly higher, with statistical significance (p<0.01),and With the cycle of CHF rats extended, the UⅡlevels increased, CHF2-week group(21.38±4.93) ng/ml, CHF4-week group(47.63±14.24) ng/ml, CHF8-week group(119.58±24.91) ng/ml, Significantly higher than normal group(5.20±1.25) ng/ml, Comparison between groups were significant or significant higher statistically (p<0.05 or p<0.01).4. Determination of hemodynamics:compared with the NS group, CHF group LVSP,±LVdp/dtmax were significantly decreased (p <0.01), LVEDP increased(p<0.01).5. Plasma brain natriuretic peptide in rats:BNP level in model 2 weeks group was higher than the normal control group, and model 4 weeks and 8 weeks groups were significantly higher than the normal control group.6.Correlation analysis:(1) plasma UⅡlevels and plasma BNP levels were significantly positively correlated, r=0.912(P<0.01); (2)plasma UⅡlevels and left ventricular systolic pressure (LVSP) were significantly negatively correlated, r=-0.800(P<0.01); (3) Plasma UⅡlevels and maximal rising rate of left ventricular systolic pressure (+dp/dt max) were significantly negatively correlated, r=-0.894(P<0.01).conclusions:1.Isoproterenol, Subcutaneous injection, could cause clearly injury of myocardium pathologically, Hemodynamics:the existence of congestive heart failure was also confirmed,2. UⅡlevels in plasma and heart function were negative correlation in model group rats,3. plasma UⅡcontent and plasma BNP content were positively correlated in model group rats,4. plasma UⅡcontent changes may be used as objective indicators in severity of heart failure.5. CHF TCM syndrome is closely related with the level of UⅡ. |
