Objective:To study the expression of Wnt5amRNA in AL(acute leukemia) of child, and compare the level of Wnt5amRNA in the Mononuclear cells of kinds of AL patient's marrow, investigate the relationship between Wnt5amRNA expression and response to treatment with AL, Provid a new molecular marker and theoretical foundation for early to judge the clinical outcome of leukemia, predicting recurrence and individualized treatment. Methods:61 Bone marrow samples including Onset and remission from 29 patients of acute leukemia as subjects and 15 cases of non-leukemia as control group. Total RNA is extracted by Catrimox-14TM RNA isolation kit. After reverse transcription, do fluorescent real-time quantitative PCR with SYBR Green I. Specificity of amplified products is double affirmed by melting curve and gel electrophoresis, The Wnt5amRNA relative expression is calculated by the way of 2-ΔΔCT and by ABL as an internal reference. Statistical analysis of relative expression of Wnt5a in different periods of AL. Results:1,the Wnt5amRNA's expression of initial treatment group with acute leukemia (0.379±2.867) is lower than non-leukemia group (22.627±35.134) very much (P=0.004).Among the ALL (acute lymphoblastic leukemia),AML (acute myeloid leukemia) of expression are 0.467±3.578 and 0.309±1.624,significantly lower than control group, has statistically significant.2,the Wnt5amRNA's expression of initial treatment group with ALL is significantly lower than ALL-CR group (3.242±118.393) (P=0.033); and the expression of initial treatment group with AML is also significantly lower than AML-CR group (1.763±30.076) (p=0.032);3,The expression of AL-CR has no significant difference than non-leukemia group (P=0.784), among the expression of ALL-CR group and AML-CR group has no significant difference than non-leukemia group, has not statistically significant. Conclusion:1,the expression of initial treatment group with AL(ALL,AML) is lower than non-leukemia group, however, has restoration at catabasis. Down-regulated expression of Wnt5a at AL has a tumor suppressor gene function. Wnt5a as a tumor suppressor gene in leukemia development may have an important role.2,AL Wnt5a expression and clinical and treatment response of children is closely related. The expression of Wnt5a in the complete remission group of was significantly higher than in the untreated group, This is highly suggested that Wnt5a is a important molecular markers in AL prognosis and treatment response, and in AL prognosis and individual therapy have important clinical value.3,Loss or reduced expression of Wnt5a is a molecular indicators of Poor prognosis. Dynamic monitoring the Wnt5a gene expression of AL-CR children can help determine prognosis and predict recurrence. Wnt5a gene can be used to monitor MRD in children with acute leukemia.
