| Object:the difference of gene experssion in Foxl 2 in hepatic stellate cells of mouse, Smad 3,which is the impontant marker in transforming growth factor-beta/Smad signing pathway, and collagen typen I,the prodution of hepatic fibrosis,the Role and infulence of forkhead L2 in transforming growth factor-beta/Smad signing pathway meditated activation of hepatic stellate cells is detected by RT-PCR. Method:after the culture of hepatic stellate cells,the TGF-beta/Smad cell signal pathway was activated by transforming growth factor-beta (5ug/L),then divided into 18 groups,RNA was extracted from every group.FOXL2, Smad3, collagen I were amplifed by their primers in every groups.Electrophoretic GEL was analysis by 1D-GEL software.3 kinds of data were analysis by linear correlation.Results:l,the OD vaule of FOXL2 showed no correlation between the expression of FOX L2 and TGF-beta/Smad signal pathway 2,the expression of FOX L2 was nothing in 16 groups.2 groups had a little expression.it means Fox L2 played no role in activaion of hepatic stellate cell.3:it was proved by the experiment that the production of TGF-beta/Smad was collagen type I.but the OD values of Smad3 was weaker than collagen type I,so the possibility of other cell signal pathway was not excluded by these OD value.Conclusion:1. there are no crosstalking between Fox L2 and TGF-beta/Smad cell signal pathway in hepatic stellate cell; 2.it shows Fox L have no relationship with TGF-beta/Smad cell signal pathway. |
