It may provide theoretical evidence for whether Id-1 is a new target for preventing and treating esophageal carcinoma. It may establish fundament for highlighting mechanism and progression of esophageal carcinoma as well.Methods: Id-1mRNA and protein expression in esophageal carcinoma and paired adjacent esophageal mucosa were detected in 30 cases, and then analyzed the relation between their expression and clinic charactors; Id-1 protein were detected by immunohisochemistory in 80 cases, and studied the relationship between Id-1 protein and the prognosis through analysing the relation between their expression and clinic charactors. Human esophageal carcinoma cell line Eca109 was conventional cultured. Effect of HCPT on cell migration and cell chemotaxis was evaluated by Wound Healing assay and Boyden cabin; further more detected the Id-1 expression in cell line by immunocytochemistry and western blot to investigate whether Id-1 was a target of HCPT on esophageal carcinoma therapy.Results:1 Id-1 expression in esophageal carcinoma and paired adjacent esophageal mucosa and the relationship between clinic charactersId-1 mRNA expression shown by RT-PCR: higher in carcinoma than in adjacent esophageal mucosa group (P<0.05); higher in well differentiation than in moderate and poor differentiation(P<0.05); there was a negative correlation with pathological grading of the tumor(r=-0.559); there was not correlation with lymphatic metastasis, depth, clinical stage, gender or age(P>0.05). Id-1 protein expression shown by Western blot: higher in carcinoma than in adjacent esophageal mucosa group (P < 0.05); higher in well differentiation than in moderate and poor differentiation(P<0.05); there was a negative correlation between Id-1 protein and pathological grading (r=-0.559); higher in lymph node metastasis cases than in non-lymph node metastasis cases(P<0.05); there was not correlation with depth, clinical stage, gender or age (P>0.05). Id-1 protein expression shown by Immunohistochemistry: higher in carcinoma than in esophageal mucosa tissue(χ2=12.325, P<0.05); there was a negative correlation between Id-1 protein and pathological grading of the tumor(r=-0.285); higher in lymph node metastasis cases than in non-lymph node metastasis cases(P<0.05); there was not correlation with depth, clinical stage, gender or age (P>0.05).Kaplan-Meier survival analysis suggested that the expression of Id-1 protein was correlated with the prognosis of esophageal carcinoma (P<0.05); Multivariate analysis of Cox model showed that the expression of Id-1 influece the prognosis(P<0.05), In Id-1 higher expression cases, the risk of death is 2.314 times than in lower expression cases. Thus Id-1 expression may be a significant prognostic indicator.2 The effect of HCPT on human esophageal carcinoma cell line-Eca109 and regulation of HCPT on Id-1 proteinWound Healing assay showed that the inhibition of cell migration increased with the HCPT concentration increasing; Boyden cabin showed that the migration of cells through the transwell significantly reduced with the HCPT concentration increasing; Immunocytochemistry and western blot all showed that the expression level of Id-1 protein decreased with the HCPT concentration increasing.Conclusions:1 Id-1 mRNA and protein expression is higher in esophageal carcinoma than in esophageal mucosa, which inhibits that Id-1 plays an important role in the carcinogenesis of esophageal carcinoma.2 The higher expression of Id-1 protein is correlated with lymph node metastasis and pathological grading of esophageal squamous cell carcinoma, which inhibits that Id-1 may be involved in malignant development through infiltration and metastasis progression.3 The higher expression of Id-1 protein inhibits lower survival rate, which may be helpful in treating esophageal squamous cell carcinoma and judging the prognosis of patients.4 HCPT could obviously inhibits the motility and chemotaxis of cultured Eca109 cell line, which existed a relationship with the dosage.5 The expression level of Id-1 protein decreased with the HCPT concentration increasing, which indicates the treating mechanism of HCPT may be related with down-regulation of Id-1 protein. | | Keywords/Search Tags: | esophageal squamous cell carcinoma, Id-1, HCPT, prognosis, Eca109 cell line | PDF Full Text Request | Related items |
