Objective: In this study, rats with behavioral changes in substantia nigra DAT, VMAT2 mRNA expression as well as serum N0, SOD, MDA assay to study the Shujin Jiedu Prescription of 6 - OHDA -induced rat model of preventive and therapeutic effects, thus revealing the Shujin Jiedu Prescription effects of the possible mechanisms of PD, for traditional Chinese medicine treatment of PD provide some experimental basis and clinical reference .Methods: Experiment selected healthy adult SD rats 100, male and female in half, Using the right brain stereotactic SNc and the VTA injection established model rat , to 6 -OHDA model was successful 60 PD rats and sham operation group 12 rats were included in the experiment, according to a stratified random sub-grouping method sham operation group, model group, LD group, SJS group, SJM group, SJL group. Behavior in rats were observed ; using real time quantitative PCR detection of rat substantia nigra DAT, VMAT2 mRNA expression; Serum using kits detect N0, SOD, MDA content.The experimental results:1 The impact of the Shujin Jiedu Prescription on PD rats behavior study Compared with those before treatment and after treatment, SJM group, SJL group and LD group start-up time, the average speed and duration were improved (P<0.05); Compared with model group and SJS group after treatment, LD group, SJM group and SJL group marked improvement in the start-up time, average speed, with statistically significant (P<0.05); with the model group and SJS group after treatment, SJM group and SJL group continued to have significantly improved the time, with statistical significance (P<0.05).2 The impact of the Shujin Jiedu Prescription on substantia nigra of PD rat DAT and VMAT2 mRNA expressionThis article observed mRNA expression of the DAT gene, VMAT2 gene in substantia nigra and the Shujin Jiedu Prescription in the substantia nigra of the mechanism of apoptosis. DAT test results showed that sham-operation group and model group, SJS group, significant differences (P<0.01); SJM group, SJL group and model group, SJS group, with statistical significance (P<0.05); VMAT2 test results showed that sham-operated group, SJM group, SJL group and model group, SJS group, was statistically significant (P<0.05).3 The impact of the Shujin Jiedu Prescription on PD rat serum N0, SOD, MDACompared with the sham-operated group , the serum NO of PD rats was significantly higher (P<0.01), SOD activity significantly decreased compared (P<0.01), MDA rose significantly higher(P<0.01), with significant differences; The PD rats after treatment in all the modeling group comparison, SJM group and SJL group, LD group SOD activity was significantly higher than SJS group and model group (P<0.05), and NO, MDA levels were lower than the model group and SJS group (P<0.05).Conclusion:1 Traditional Chinese medicine, the middle and high dose group and western group significantly improved both the symptoms of PD in rats, while the low dose group had no significant effect; while the Shujin Jiedu Prescription to improve the symptoms of PD in rats has a dose-effect relationship.2 Model rat compared with the sham-operated group of DAT, VMAT2 mRNA expression was significantly reduced. The Shujin Jiedu Prescription middle and high dose group DAT, VMAT2 gene amplification products were significantly higher than PD model group, western medicine group, traditional Chinese medicine low dose group, indicating the Shujin Jiedu Prescription of Parkinson's disease substantia nigra in rats DAT, VMAT2 gene increases role and can ease from 6-OHDA induced DAT, VMAT2 expression lower than western medicine and Chinese medicine low dose group, and the drug dose-effect relationship exists.3 6-OHDA rat serum after modeling there is the situation of excessive oxidative stress, free radical scavenging capacity decreased. The Shujin Jiedu Prescription can antagonize the neurotoxicity of PD rats, N0, improve anti-free radical capacity, to a certain extent, can improve the anti-oxidation disorders. Traditional Chinese medicine in improving oxidative stress and over-dose-effect relationship exists.
