Objective: (1) To examine the expression level of visfatin mRNA in subcutaneous adipose tissue (SAT) , omental adipose tissue (OAT) and plasma visfatin and its relationship with related factors in Uygur obesity subject. (2) Investigate visfatin gene expression during the period of 3T3-L1 preadipocyte differentiation. Methods: 56 Subjects were divided into obesity group and non-obesity group according to BMI①RT-PCR was used to investigate the expression of visfatin mRNA from SAT and OAT②Measurement of anthropometric and metabolic parameters in uygur subjects. and calculation of BMI, WHR.③3T3-L1 preadipocyte were cultured invitro and were induced to differentiated into the matured adipocytes. The differentiation of adipocytes was assessed through detecting expression of visfatin by RT-PCR and Western blot and oil red O staining. Results:①Expression level of visfatin mRNA in obesity group is higher than the non-obesity group, but no significant difference. plasma visfatin concentration was increased significantly in obesity patients (P<0.05) .②Correlation analysis showed that the expression level of visfatin mRNA in OAT was correlated with BMI (r=0. 26, P<0. 05) and TG (r=0.55, P<0.05) and the expression level of visfatin mRNA in SAT was significantly correlated with TG (r=0.54, P<0.05) .③Plasma visfatin concentration was positively correlated with weight (r=0. 38, P<0.05) , waist circumference (r=0.33, P<0.05) , hip circumference (r=0.27, P<0.05) , but was negatively correlated whith HDL-C (r=-0.27, P<0.05) .④During the differentiation phase of 3T3-L1 adipocyte, the expression of visfatin mRNA is increased, and the mRNA expression of visfatin gene was kept higher level and remained unchanged in the telophase. Expression level of visfatin mRNA from the sixth day to twelfth day is higher than from 0 to fourth day, from the tenth to twelfth day the expression of visfatin is higher than the sixth day, the twelfth day expression of visfatin is higher than the eighth day.⑤. The results of western blot show that as the adipocyte goes to mature the expression of protein is increased. Conclusion: Visfatin may be related to the development of obesity and lipid metabolism in Uygur. Expression of visfatin mRNA and protein increases during the differentiation of preadipocytes, visfatin may be involved in lipid synthesis and accumulation.
