| Objective:To study the effect and mechanism of the traditional fermented cheese whey (TFCW) in Xinjiang on rats with experimental hyperlipidemia. To separated the bioactive peptides with ACE inhibitory or antioxidant.Methods:The hyperlipidemia was induced by feeding rats with higncholesterol diet.Rats wererandomly divided into 5 group,and treated with cheese whey(50,100mg/kg),or pravastatin(20 mg/kg).the levels of serum TC,TQHDL-C,LDL-Cweremeasure.Nitric(NO),hydroxylradical(·OH),lecithin cholesterol acyltransferase(LCAT) of the rats in different groups were detected. TFCW is used as raw material, We separated TFCW by 30kDa, lOkDa and 5kDa membrane ultrafiltration and extracted by Sephadex G-15 column.The optima conditions of papain hydrolysis was obtained by orthogonal with with the ACE inhibitory ratio in vitor,DPPH·,·OH,O2-·scavenging ratio as index. Results:Compared with the model group,the two dose of TFCW showed obvious reduction in the contents of TC,TG,LDL-C,raise HDL-C.The results also showed raise the contedt of NO,strengthen the scavenging capacity of·OH,and enhance the activity of LACT. Under this conditions were 40℃, O.1Mpa, three peptide were extracted with 30-lOkDa,10-5kDa,≤5kDa by ultrafiltration.This peptides were separated to five components by Sephadex G-15 column. F4 which has a high rate of ACE inhibition, was 56.45%, and the relative molecular weight distribution in the 1071-724Da; F5 has a high free radical scavenging capacity, DPPH·,·OH, O2-·inhibition rate was 59.06%,62.06%,29.51%, the relative molecular weight distribution in the 645-500Da. Conclusion:TFCW has adjusted the blood lipids.The mechanism may be related to the incredsing activity of LCATand the savenging capacityof free radical.The peptide of TFCW has strongly ACE inhibitory activity and antioxidant activity in vitro. |
