Flow cytometry analysis(FCM) was taken to measure the Cell cycle. Transwell chamber was used to determine the invasion ability of the cell lines. Micropipette aspiration technique was taken to measure the adhesion force of KAI1-HNE-1 cells.Results①When the concentration of the adenovirus was 30 MOI,the transferred rate of HNE-1 cell line was above 92%.②MTT showed the cell proliferation was inhibited compared to the contrast group(p <0.05).The inhibition rate was up to 30%.③The FCM of the KAI1-HNE-1 cell line showed that the cell cycle was blocked in the G0/G1 phase.④The invasion assay demonstrated that the number of HNE-1 cells which crossed the membrane in KAI1 transfection group(52.5±4.2) was lower than that in the non-transfected group(167.4±3.5).The difference was statistically significant(p<0.05).⑤The homogeneity adhesion of KAI1-transfected HNE-1 cells group was increased(p<0.05).Conclusion The results demonstrated that KAI1 can inhibit the cell proliferation of the HNE-1 cell line and the possible mechanism was arrested the cell cycle at G0/G1 checkpoint.The invasion ability of HNE-1 cells was reduced by KAI1 gene,the adhesion of KAI1-HNE-1 cells were increased. | | Keywords/Search Tags: | KAI1, HNE-1, proliferation, invasion, metastasis | PDF Full Text Request | Related items |
