| Objective:To study the effect of mitomycin C for the rabbit ears incision outside the pathologic scar, scar thickness,morphology, proliferation of fibroblasts and collagen fibers and other aspects,so as to provide theoretical and experimental basis for the preventing and topical mitomycin the treatment of skin proliferation of scar formation.Methods:After the success in producting the rabbit ears scar model, with 0.05mg/ml and 0.1mg/ml of mitomycin C solution of rabbit ears wound dressing,and using the blank group and the triamcinolone group and control group,the same method wound dressing,local reserves, until wound healing,surgical procedures and are strictly aseptic dressing change,the growth observed with color doppler scan scar thickness,was cut to do biopsy scar, using Van Gieson stain,elastic fibers-collagen combined staining of collagen fiber growth,HE staining of fibroblast proliferation,and according to each HE section of random three high magnification view,and counted the number of fibroblasts,counting results and the scar thickness of statistics were used spss14.0 software for image analysis and statistical analysis.Result Morphological and histological results of general observation:the naked eye observation:color of the control group scar is depth than triamcinolone group,0.1mg/ml and 0.05mg/ml group group,and texture hard,surface unsmooth.Triamcinolone group,0.1 mg/ml group scar' color are lighter. HE staining observed in control group scar tissue sections marked capillary proliferation,large fibroblast nuclears,dense hyperplasia,in full-thickness dermal collagen disordered.Triamcinolone group and 0.1mg/ml group are thin scar tissue,surface flat,level order, fibroblasts significantly reduced.Van Gieson staining showed that the collagen fibers of saline group (control group) were irregularly arranged, local formation of vortex, collagen fibers of triamcinolone group and 0.1mg/ml group decreased,tight and orderly. Elastic fibers-collagen fibers had no obvious co-staining elastic fibers,but the level of each group were markedly higher than triamcinolone group and 0.1mg/ml group disorder, similar to the other with Van Gieson staining.Color Doppler scanning scar thickness between 0.1mg/ml group and triamcinolone group were no statistically significant(P> 0.05).But the control group compared with the 0.1mg/ml group and the triamcinolone group,resulting is significant statistical difference(P<0.01).Conclusions:Topical mitomycin C can effectively reduce the rabbit ear scar formation,as long as the control at a reasonable level of low concentration,it would not cause local necrosis and the other side effects. Scar thickness from the color Doppler scan statistics:0.1mg/ml MMC concentrations inhibited the growth of scar with triamcinolone results were similar,0.1mg/ml concentration group,the effect of mitomycin C significantly inhibited the growth of scar better than the control group and 0.05mg/ml concentration.High magnification view from the fibroblast count statistics:0.1mg/ml MMC concentrations inhibit the proliferation of fibroblasts and the effect of triamcinolone was similar,0.1mg/ml MMC concentrations inhibit the hypertrophic scar fibroblasts cells better than the control group and 0.05mg/ml concentration. |
