Objective:The esophagus carcinoma is one of the common malignant tumors in humanbeings.Our country is the prone area to the esophagus carcinoma and more than a half of esophagus carcinoma patients in the world are Chinese according to preliminary estimates.There are about 150 thousand patients dying of esophagus carcinoma every year in our country,almost one fourth of all the patients dying of malignant tumors.As the development of medicine,the therapeutic efficacy of the esophagus carcinoma in the near future is advancing gradually but how to prevent the recurrence after the operation and raise the long term survival rate are still the tough problems in the medicine field so it is very significant to study the genesis,infiltration and metastasis mechanism of the esophagus carcinoma. This experiment is to study the correlation of the CD24,CD105 and NF-ΚB expression in the esophagus squamous cell carcinoma and the normal esophagus mucous membrane tissues by testing the expressions and the quantity of the CD24,CD105 and NF-ΚB in the the esophagus squamous cell carcinoma and the normal esophagus mucous membrane tissues and furthermore to supply the theory evidence for the research of the genesis,infiltration and metastasis mechanism of the esophagus carcinoma and the prevention methods of the esophagus carcinoma genesis and development.Methods:Forty tumor and normal tissue(the distance between the superior incisal edge of the sample and the edge of the tumor is more than 5cm and no invasion by the tumor) samples of this study were obtained from the esophagus carcinoma patients undergoing surgical resection in the cardiothoracic surgery department of the Fourth affiliated hospital of HeBei Medical University from February to July of 2009.All the patients had no radiotherapy or chemotherapy before the operation.The samples of the tumor are all squamous cell carcinoma by the pathology examination. S-P method of Immunohistochemistry was used to detect the expression of the CD24,CD105 and NF-ΚB in the the esophagus squamous cell carcinoma and the normal esophagus mucous membrane tissues of the forty esophagus carcinoma patients undergoing surgical resection and to study the correlation of them in the gender,years of age,the size of the tumor,the level of the differentiation,whether invading the external membrane,the clinical staging and the lymph node metastasis by statistical analysis of software SPSS17.0.Results: 1 The CD105-positive expression rate is higher in tumor tissues than that in the normal esophagus mucous membrane tissues and the difference is obvious The CD24-positive expression rate is higher in tumor tissues than that in the normal esophagus mucous membrane tissues and the difference is obvious(χ2=14.066, P=0.00). The NF-ΚB-positive expression rate is higer in tumor tissues than that in the normal esophagus mucous membrane tissues and the difference is obvious(χ2=19.527, P=0.00).2 Between the StageⅠandⅡand StageⅢesophagus squamous cell carcinoma tissues,the numerical values of MVD marked by CD105 antibody are 32.40±7.00 and 42.50±3.10 respectively and the difference between them has statistical significance(P<0.05). Between the poorly differentiated esophagus squamous cell carcinoma tissues and the moderately and well differentiated esophagus squamous cell carcinoma tissues,the numerical values of CD105-positive expression are 41.83±5.60 and 33.48±7.34 and the difference has statistical significance(P<0.05). Between the esophagus squamous cell carcinoma tissues with lymph node metastasis and that without lymph node metastasis,the numerical values of CD105-positive expression are 35.09±8.25 and 34.71±6.95 and the difference has no statistical significance(P >0.05). In the aspect of the gender, the numerical values of CD105-positive expression are 35.09±8.25(male) and 34.71±6.95(female)and the difference has no statistical significance(P >0.05).In the aspect of tumor size(≥5cm or <5cm) the numerical values of CD105-positive expression are 37.36±7.95 and 34.00±7.44 and the difference has no statistical significance(P >0.05). In the aspect of the age (≥60 yeares or <60 years) the numerical values of CD105-positive expression are 34.58±6.75 and 35.44±9.01 and the difference has no statistical significance(P >0.05). Between the esophagus squamous cell carcinoma tissues with external membrane invasion and that without external membrane invasion,the numerical values of CD105-positive expression are 38.00±5.01and 31.07±8.47 and the difference has statistical significance(P<0.05).3 Between the StageⅠandⅡand StageⅢesophagus squamous cell carcinoma tissues,the CD24-positive expression rate is 46.7% and 80.0% respectively the difference has no statistical significance(P >0.05). Between the poorly differentiated esophagus squamous cell carcinoma tissues and the moderately and well differentiated esophagus squamous cell carcinoma tissues, the CD24-positive expression rate is 85.7 % and 30.3% and the difference between them has statistical significance(P<0.05). Between the esophagus squamous cell carcinoma tissues with lymph node metastasis and that without lymph node metastasis, the CD24-positive expression rate is 81.8% and 27.6% and the difference between them has statistical significance(P<0.05). In the aspect of the gender, the CD24-positive expression rate is 60.8%(male) and 47.1%(female) and the difference has no statistical significance(P >0.05). In the aspect of tumor size(≥5cm or <5cm) the CD24-positive expression rate is and the difference has no statistical significance(P >0.05). In the aspect of the age (≥60 yeares or <60 years) the CD24-positive expression rate is 58.3% and 56.3% respectively and the difference has no statistical significance(P >0.05). Between the esophagus squamous cell carcinoma tissues with external membrane invasion and that without external membrane invasion, the CD24-positive expression rate is 72.0% and 20.0% and the difference between them has statistical significance(P<0.05).4 Between the StageⅠandⅡand StageⅢesophagus squamous cell carcinoma tissues,the NF-ΚB-positive expression rate is 53.3% and 80.0% respectively and the difference has no statistical significance(P >0.05). Between the poorly differentiated esophagus squamous cell carcinoma tissues and the moderately and well differentiated esophagus squamous cell carcinoma tissues, the NF-ΚB-positive expression rate is 100% and 51.5% the difference has no statistical significance(P >0.05). Between the esophagus squamous cell carcinoma tissues with lymph node metastasis and that without lymph node metastasis, the NF-ΚB-positive expression rate is 90.0% and 16.7% and the difference between them has statistical significance(P<0.05). In the aspect of the gender, NF-ΚB-positive expression rate is 60.8%(male) and 58.8% (female) and the difference has no statistical significance(P >0.05). In the aspect of tumor size(≥5cm or <5cm) NF-ΚB-positive expression rate is 100% and 44.8% and and the difference between them has statistical significance(P<0.05).In the aspect of the age (≥60 yeares or <60 years) NF-ΚB-positive expression rate is 54.2% and 62.5% respectively and the difference has no statistical significance(P >0.05). Between the esophagus squamous cell carcinoma tissues with external membrane invasion and that without external membrane invasion, the NF-ΚB-positive expression rate is 76.0% and 33.3% and the difference between them has statistical significance(P<0.05).5 The numerical value of CD105 is 37.91±5.66 in the samples with the CD24-positive expression and 31.28±8.27 in the negative ones and the difference has statistical significance(P<0. 05).6 The numerical value of CD105 is 37.54±5.96 in the samples with the NF-ΚB-positive expression and 31.00±8.35 in the negative ones and the difference has statistical significance(P<0. 05).7 The outcome of the Spearman correlation analysis indicates that the positive expression rate of the CD24 and NF-ΚB in the esophagus squamous cell carcinoma tissues is obvious positive correlation(r=0.595,p=0.00<0.05) .Conclusion:1 The positive expression of CD105, CD24 and NF-ΚB is much higher in the esophagus squamous cell carcinoma tissues than that in the normal esophagus tissues,which indicates that the high expression of CD105, CD24 and NF-ΚB may has some relation with the genesis and development of the esophagus carcinoma.2 This experiment used anti-CD105 monoclonal antibody to test the MVD of the new vessels in the esophagus squamous cell carcinoma tissues and the outcome indicated that the MVD in the esophagus squamous cell carcinoma tissues has nothing to do with the age,gender,the lymph node metastasis and the size of the tumor but correlates with the invasion depth,the level of differentiation and the clincal staging positively ,which shows that MVD can be use as an index to justify the invasion depth,the level of differentiation and the clincal staging of the esophagus squamous cell carcinoma tissues.3 The CD24-positive expression rate in the lymph node metastasis, the level of differentiation and the external membrane invasion of the esophagus squamous cell carcinoma tissues has statistical significance but doesn't have statistical significance in the gender,the size of the tumor,the age and the staging of the tumor,which indicates that the high expression of the CD24 has relation with the the lymph node metastasis, the level of differentiation and the external membrane invasion of the esophagus squamous cell carcinoma tissues.4 The NF-ΚB-positive expression rate in the lymph node metastasis, the size of the tumor and the external membrane invasion of the esophagus squamous cell carcinoma tissues has statistical significance but doesn't have statistical significance in the level of differentiation,the gender, the age and the staging of the tumor,which indicates that the high expression of the NF-ΚB has relation with the malignant level of the tumor,the lymph node metastasis and the external membrane invasion of the esophagus squamous cell carcinoma tissues.5 CD105, CD24 and NF-ΚB have positive relation,which indicates that the expression of CD105, CD24 and NF-ΚB is coincident.Test CD105, CD24 and NF-ΚB at the same time and the outcome can be used as an objective index to evaluate the invasion and metastasis ability of the esophagus squamous cell carcinoma and to provide base for the treatment after operation and is significant to the prognosis.
