Objective: Ulcerative Colitis is one of the refractory digestive diseases. Traditional Chinese Medicine treatment has abundant experience to cure the disease. Lan Yin Feng Yang Hua Zhuo Jie Du decoction (LYFY) is a experiential prescription in clinical of my professor.Through the establishment of rat model of ulcerative colitis to observe the influence on the expression of nuclear factor-kappa B and inhibitor ofκB which are in rat ulcerative colitis tissue. The study explains the possible role of the treatment of UC mechanism of the therapy in order to provide experimental evidence.Methods: 60 healthy Wistar rats of clean grade were randomly divided into five groups with 12 rats in each group: the normal group (N group) , the model group (M group), the sulfasalazine group (SASP group), the high-dose LYFY ( HD group), the low-dose LYFY (LD group). In addition to the normal group, the other four groups were controlled in order to make Ulcerative Colitis animal models by the way of TNBS/ethanol combinative method. The first 3 days after modeling, the normal group and model group fed with normal saline.And the other group rats were treated for 14 days. We observed the changes of activity, body weight, general changes in stool, etc.; calculated ulcer in each group the number, the evaluation of rat colon tissue inflammation, congestion and edema degree; using immunohistochemical staining of rat colon mucosa IκB-α, NF-κB expression. Efficiency evaluation based on the observed results, analyzing LYFY on IκB-αand NF-κB effects.Results:1 The general stateCompared With the normal group, model rats lack of energy, activity decreased extrados, hair loose, dull yellowish fur, decreased appetite, Diarrhea will be bloody, or mucus, shape increasingly thin. One week after drug treatment, HD group and SASP group of rats loose stools or soft then gradually stop, diet, activity been increasing gradually re-shiny hair; LD rats had some improvement, but less improved than HD group.2 The general observation of colonic tissueCompared With the M group, the SASP group and HD group have a few of ulcers.The number of ulcer on M group, HD group,SASP group and LD group were (2.82±0.98), (1.17±0.94),(1.82±0.98)and (2.00±1.04),HD group and SASP group were significantly decreased compared to M group (p< 0.05). HD group and SASP group have not remarkable difference, and the differences have no statistically significant difference (p﹥0.05). LD group and M group have not remarkable difference, and the differences have no statistically significant difference (p﹥0.05).The index of injury of M group,HD group,SASP group and LD group were (3.73±0.90),(2.58±1.24),(2.64±0.92) and (3.25±1.42),HD group and SASP group were significantly decreased compared with M group (p<0.05). HD group and SASP group have not remarkable difference, and the differences have no statistically significant difference (p﹥0.05). LD group and M group have not remarkable difference, and the differences have no statistically significant difference (p﹥0.05).3 The observed under optical microscopeThe degrees of inflammation of HD group was significantly decreased compared with M group (p<0.05), but SASP group was not(p﹥0.05). HD group and SASP group have remarkable difference, and the differences have statistical meaning (p<0.05). LD group and SASP group have not remarkable difference, the differences have have no statistically significant difference (p﹥0.05).The M group rats mucous layer missing, congestion and edema in the submucosa can be seen, and inflammatory cell infiltration, HD group level of mucosal edema and congestion, were significantly lighter than M rats (P <0.05); while the SASP group only in improving the degree of congestive heart statistically significant (P <0.05).4 The test results of IκB-αand NF-κB p65The expression level of IκB-αin M group (20.05士1.77)% is higher than the level of N group (10.33士1.36)% (p<0.05). The expression level of IκB-αin HD group,SASP group and LD group were (11.41士1.46)%,(13.13士1.67)%,(13.28士1.55)%.The treated groups were significantly decreased compared with M group (p<0.05). HD group and SASP group have remarkable difference, and the differences have statistical meaning (p<0.05). HD group and LD group have remarkable difference, and the differences have statistical meaning (p<0.05). SASP group and LD group have no significant differences(p﹥0.05). Compared with the expression of IκB-αin N group, HD group has no significant differences(p﹥0.05),but SASP group and LD group have remarkable difference (p<0.05).The expression level of NF-κBp65 in M group (19.42士1.24)% is higher than the level of N group (8.70士1.11)% (p<0.05). The expression level of NF-κBp65 in HD group, SASP group and LD group were(9.12士1.36)%,(10.35士0.95)% and (10.50士1.02)%.The treated groups were significantly decreased compared with MG(p<0.05).HD group and SASP group have remarkable difference, and the differences have statistical meaning(p<0.05). HD group and LD group have remarkable difference, and the differences have statistical meaning (p<0.05). SASP group and LD group have no significant differences(p﹥0.05). Compared with the expression of NF-κBp65 in N group, HD group has no significant differences(p﹥0.05),but SASP group and LD group have remarkable difference (p<0.05).Conclusion:1 LYFY can improve the general situation in UC rats, reducing the number of rats with colon mucosa ulcers, improve colonic mucosa hyperemia, edema and other pathological changes. High doses of traditional Chinese medicine, compared with sulfasalazine in reducing the number of ulcers and improve the damage integral has the same effect; while improving the degree of inflammation and reduce IκB-α, NF-κB expression level is concerned, the role of high doses of traditional Chinese medicine is better than SASP.2 LYFY can reduce the level of IκB-αand NF-κB expression, promote a combination of both, thereby reduce the activity of NF-κB to limit the release of cytokines, reduce intestinal inflammation and immune response,and promote ulcer healing. Experiments show that LYFY has a good therapeutic effect in UC rats copied by the method of TNBs / ethanol co-modeling. |
