| Objective:Ulcerative colitis(UC), is an idiopathic chronic inflammatory bowel disease. Its main clinical manifestations are diarrhea, bloody mucopurulent stools, abdominal pain and tenesmus. Respecting the disease is difficult to cure,we have been lacking of effeetive remedies for treating it. Therefore, finding effective prevention and treatment of drug have also been a research hotspot. Fortunately,we have a long history and rich experimence to treat UC by using traditional Chinese medicine. LanYinFengYangHua- ZhuoJieDu Decoction which has significant effect and few side effect is a common dose to cure UC. Through the experiment,we could observe Lan Decoction's effects on UC and demonstrate the immune functional mechanism.Methods:In the experiment,60 Wistar rats were randomly divided into five groups,every group had 12 rats. namely the normal control group(NG), the model control group (MG),the sulfasalazine group(SASP),the Lan Decoction high-dose group (HD), the Lan Decoction low-dose group(LD). Except the normal control group ,the other four groups were made through the 2,4,6 - Trinitrobenzene sulfonic acid (TNBS) / ethanol .These three groups were treated by SASP,high-dose,low-dose of Lan Decoction .The subject was studied by three aspects: First, observation of Lan Decoction on the general state and colonic pathological morphology;second, observation of Lan Decoction on T-lymphocyte subsets;Third, observe the Lan Decoction on cytokines IL-8 and IL-10.Results:The general condition,tissue damaged seore and pathological damaged State The study showed that the weight of rats in Model group, Lan Decoction low-dose group, Lan Decoction high-dose group, SASP group were lower than that in normal group(P<0.05);in normal group,Chinese medicine low-dose group,Chinese medicine high dose group, SASP group were higher than that in Model group, (P<0.05);and the weight in Lan Decoction high-dose group were higher but low-dose group were lower than SASP.(P<0.01)The colon tissue damaged seore in model group was higher than that in the high-dose,low-dose group of Lan Decoction and SASP group(P<0.05);and the score in Lan Decoction high-dose group were lower but low-dose group were higher than SASP.(P<0.01) There was more difference in gland arrangement being regular or not,mucosa hyperemia and inflalmmation cell infiltration and so on in each group.2 The T-lymphocyte subsetsThe level of CD4+ in each group were not statistically significant (P> 0.05).The level of CD8+in Model group, Lan Decoction low-dose group, Lan Decoction high-dose group,SASP group were lower than that in normal group(P<0.05);in normal group,Lan Decoction low-dose group,Lan Decoction high-dose group, SASP group were higher than that in Model group, (P<0.05);and in Lan Decoction high-dose group were higher but low-dose group were lower than SASP.(P<0.01)The ratio of CD4+/CD8+in Model group, Lan Decoction low-dose group, Lan Decoction high-dose group,SASP group were higher than that in normal group(P<0.05);in normal group,Chinese medicine low-dose group,Chinese medicine high dose group, SASP group were lower than that in Model group, (P<0.05);and in Lan Decoction high-dose group were lower but low-dose group were higher than SASP.(P<0.01)3 The cytokinesWith the normal control group, serum IL-8 levels compared with Lan Decoction high-dose group showed no statistical significance (P> 0.05).IL-10:The level of IL-10 in Model group, Lan Decoction low-dose group, Lan Decoction high dose group, SASP group were lower than that in normal group(P<0.05);in normal group,Lan Decoction low-dose group,Lan Decoction high-dose group, SASP group were higher than that in Model group, (P<0.05);and in Lan Decoction high-dose group were higher but low-dose group were lower than SASP.(P<0.01)Conclusion:1 This model was suecessful.2 LanYinFengYangHuaZhuoJieDu Decoction depress the CD8 + count, improve CD4+ / CD8+ ratio.3 Lan Yin-Feng Yang Huazhuo detoxification may inhibit pro- inflammatory factor IL-8, enhance the anti-inflammatory factor IL-10.4 LanYinFengYangHuaZhuoJieDu Decoction has a good therapeutic effect in UC model rats. The high-dose group is more effective than SASP group. |
