Objective:Through the earlier research We observed that the general and micro-circulation changes in the mesenteric, intestinal mucosa, cortex renis, liver, gastric mucosa, deep fascia, pia mater encephali and other tissues and organs of animal models with high-voltage electrical injury. The images and date suggests that not only local skin but also remote organs result from high-tension electrical burns. The main displays of the changes of microcirculation is the change of coagulation and formation of micro-thrombus. Some research show that PAF, TXA2 and PGI2 play important roles in coagulation. The aim of experiment we designed is to further study the mechanism of microcirculation disorder by observing changes of PAF, TXA2 and PGI2 of rat models with high-tension electrical burns in the plasma, and the therapeutic effect of PTX in order to provide a theoretical basis for clinical treatment of high-tension electrical burns.Methods:1.Group:One hundred and eighty adult male SD rats weighing 330±30g g were used divided into three groups in random:high-tension electrical burns experiment group (experiment group), high-tension electrical burns PTX treatment group (treatment group) and fake high-tension electrical burns group (control group), each group has 60 rats. The time phase of these groups are divided into six different phases:ahead(T0); immediate(15min) (T1);1h(T2); 2h(T3); 4h(T4); 8h(T5), each phase has 10 rats. Number the rats according to groups and phases.2.Preparation:Weigh the rats and record the date. Rats were housed in standard laboratory conditions. The rats were given free access to water and fasted for 12h.Dispensing the experimental drug as required concentration. 3.Duplication of rat model with high-tension electrical burns:The rats were anesthetized with an intraperitoneal injection of 40mg/kg 1% sodium pentobarbital. The medial surface of the left forelimb and right thigh and thoracoabdominal of the rats were routinely prepared by shaving. Make the experimental equipments normal operation and rat prepared. Anaesthetized rats of electrical burn experiment group and PTX treatment group are shocked by the 2,000 volts electric current for 3 seconds. We immediately give rats of treatment group an intraperitoneal injection of 40mg/kg 1% PTX, physiological saline to the experiment group and the control group after cutting down the power. The progress of control group is the same as the experiment group, but there isn't electric current in the body of control group rats.4.Collection and detection:Gather blood by intracardiac penetration in living rats. Settlement for 30minutes and centrifuge with 3000r/min about 10 minutes. Abstract supernatant, and then preserve in ultra low freezer.5.The content of PAF, TXA2 and PGI2 of the rats in the plasma were measured by enzyme-linked immunosorbent assay (ELISA) at designed different time phases in early stage of high-tension electrical burns.6.Analysis of Statistics:statistically analysis all the data with SPSS 16.0. All data are expressed as the means±SD. We apply two-way analysis of variance (two-way ANOVA) among groups. The level of significance was set at *p<0.05.Results:the changes of PAF, TXA2, PGI2 and TXA2/PGI2 in the plasma.1. The levels of PAF, TXA2, PGI2 TXA2/PGI2 in the plasma have no marked difference within the control group (p>0.05).2. Compared with control group and phase To, the levels of PAF, TXA2, PGI2 and TXA2/PGI2 of the experiment group in the plasma have an marked increase (p<0.05) and they all increase from the phase T1; the levels of PAF and TXA2 of the experiment group in the plasma have obviously increase and peak at the phase T4 then decrease gradually, but still much higher at the phrase T5 than control group (p<0.05); The level of PGI2 of the experiment group in the plasma has a stable increase from the phase T1 to the phase T5;compared with PGI2, the levels of TXA2 of the experiment group in the plasma has more apparent increase from the phase T1 to the phase T4. As a result, the ratio of TXA2/PGI2 reach the peak at the phase T4. With the increasing of content of PGI2 and decreasing of content of TXA2, the ratio of TXA2/PGI2 becomes small but still higher at the phase T5 than control group (p<0.05)3. Compared with control group, The levels of PAF, TXA2, PGI2 and TXA2/PGI2 of treatment group in the plasma levels have marked increase (p<0.05), but at the same phase compared to experiment group, The levels PAF and TXA2 and the ratio TXA2/PGI2 have obvious decrease and the level of PGI2 has a apparent increase from the phase T1 to the phase T4(p<0.05). There is no marked difference at the phase T5 (p>0.05).Conclusion:1.The levels of PAF, TXA2, PGI2 and TXA2/PGI2 experiment group have obvious increase after high-tension electrical burns. They play important roles in microcirculation disorder.2.The study shows in the PTX treatment group the levels of PAF, TXA2 and TXA2/PGI2 lower than experiment group, but the levels of PGI2 higher than experiment group. At the same time in the PTX treatment group the levels of PAF, TXA2, PGI2 and TXA2/PGI2 higher than control group. PTX might be helpful in preservation and relief microcirculation disorder after high-voltage electrical burns.
