Objective To investigate the protective effects of saturated hydrogen saline on lung injury and lung fibrosis induced by paraquat(PQ) in rats.Methods 48 health male Sprague-Dawley rats were randomly divided into 3 groups:normal control group,exposure group and intervention group,16 rats in each group.For exposure group and intervention group,50 mg/kg PQ was administered to rats by gavage to set a PQ poisoned rat model,equal volume of distilled water was gavaged in control group,and an hour later 5 ml/kg saturated hydrogen saline was administered to intervention group by peritoneal injection,twice a day until executed; equal volume normal sodium was administered to other two groups by peritoneal injection.At 3 days and 21 days after contamination,the arterial partial pressure of oxygen,lung homogenate MDA,8-OhdG,wet dry weight ratio and the expression of TGF-β1 in lung tissue were measured,the HE staining of histological changes and Masson staining of collagen accumulation in lung tissue were also observed under the light microscope.SPSS 12.0 software was used for Statistical analysis,The probability of statistical differences between experimental groups was determined by Student-Keuls(SNK-q) or Kruskal-Wallis H as appropriate,Statistical differences were considered significant at p < 0.05.Results①PaO2:3 days after contamination,PaO2 in exposure group and intervention group was lower than that in control group(p < 0.01);Compared with exposure group,PaO2 in intervention group was higher(p < 0.05). 21 days after contamination, compared with control group,PaO2 in exposure group were still lower(p < 0.01),and that in intervention group was also lower(p < 0.05);PaO2 in intervention group was much higher than that in exposure group(p < 0.01).②L ung wet dry weight ratio:3 days after contamination,compared with control group,the lung Wet dry weight ratio in exposure group and intervention group was much higher than control group(p < 0.01);Compared with exposure group,the Lung Wet dry weight ratio in intervention group was lower(p < 0.05).③Lung MDA content:3 days after contamination, compared with control group, the content of lung MDA in exposure group was significantly higher(p < 0.01),and that in intervention group was higher(p < 0.05); Compared with exposure group,lung MDA content in intervention group was much lower(p < 0.01).21 days after contamination,there was no significant difference in each group(p > 0.05).④L ung 8-OHdG content:3 days after contamination, Compared with control group, the content of 8-OHdG in exposure group was significantly higher(p < 0.01),and that in intervention group was also higher(p < 0.05); Compared with exposure group,lung 8-OHdG content in intervention group was much lower(p < 0.01).21 days after contamination, there was no significant difference in each group(p > 0.05).⑤Expression of TGF-β1 in lung:3 days after contamination, compared with control group,the expression of TGF-β1 in exposure group was elevated(p < 0.01),and that in intervention group was also elevated(p < 0.05);there was no significant difference between exposure group and intervention group(p > 0.05).21 days after contamination,compared with control group,the expression of TGF-β1 in exposure group was elevated(p < 0.01),and that in intervention group was also elevated(p < 0.05); compared with exposure group,that in intervention group was significant lower(p < 0.01).⑥Pathological changes: In control group, the alveolar wall was thin and with complete structure ,alveolar space was not broadened or hyperemia,and infiltration of inflammatory cells were not observed in alveolar space,Masson stain showed that only a bit of stained green collagen fibres were found in the areas around bronchia and the interspace between alveoluses .In exposure group, the alveolar space was obviously broadened,plenty of erythrocyte and inflammatory cell infiltrated in tissue interspace and alveolar cavity ecxept apparent alveolar edema at 3d,and the red edama fluid could be seen in some alveolar spaces.At 21d there were plenty of collagen deposited,alveolar septum and part of the alveolar space were occupied by collagen and fibrin,some alveolar structures had collapsed and destroyed.Masson stain showed that plenty of stained green collagen fibres were found in alveolar septum and alveolar space.In intervention group,3 days after contamination,a certain widened alveolar space could see in some of the alveolar lung tissue,inflammatory cell infiltration could see in pulmonary interstitial and alveolar cavity,which surrounded by many normal lung tissue; 21 days after contamination, a small amount of alveolar collapsed and with a little collagen deposition,which surrounded by many normal lung tissue,Masson staining of collagen accumulation was seen in the alveolar interval, compared with poisoned group, the percentage of blue-stained area decreased (p < 0.01).Conclusion 1.Serious oxidative stress was induced by PQ intoxication in experimental animal,oxidative stress plays a very important role in paraquat poisoning. 2. Saturated hydrogen saline could alleviate lung oxidative stress injury and reduce diffuse lung injury induced by PQ;Saturated hydrogen saline could reduce lipid peroxidation and DNA oxidative damage,depress the expression of TGF-β1 level,eliminate collagen accumulation,thus,it can reducing the degree of lung fibrosis.
