Objective:Effects of triptolide on the expression of NF-κB and IκBαwhich is the inhibiting factor of NF-κB in microglial cells of the cellular model of AD were studied to explore the possible mechanism of triptolide against AD.Methods:Cultured microglial cells were induced by Aβ1-40 whose solubility is 20μl/L . Immunocytochemistry methods and image analysis were used to observe the effects of triptolide in different dose(5μg/ml,25μg/ml) on the expression of NF-κB and IκBαin microglial cells induced by Aβ1-40 in different phase(2 h,12 h,24 h).Results:After 2 hours of giving drugs,the integrated optical density(IOD) of NF-κB in every group have no significant difference.After 12 hours or 24 hours, the IOD of NF-κB in Model group was more higher than the Normal group(P<0.01).Compared with Model group, the IOD of NF-κB in Low-dose TP group was obviously decreased (P<0.01),but higher than Nomal group(P<0.01). Compared with Model group, the IOD of NF-κB in High-dose TP group was obviously decreased (P<0.01),and there is no significant difference between it and Nromal group(P>0.05).Compared with Low-dose TP group,the IOD of NF-κB in High-dose TP groups was decreased (P<0.05,P<0.01).there is a close correspondence between the expression of IκBαand the expression of NF-κB in every group.Conclusions:Triptolide can restrain the activation of NF-κB in microglial cells induced by Aβ1-40. There was positive correlation between the inhibiting effect and the dose and phase of triptolide.The fluctuant tendency of the expression of NF-κB and IκBαwas accordant. Objective:Effects of triptolide on the neuronal apoptosis in hippocampus of the cellular model of AD were studied to explore the possible mechanism of triptolide against AD.Methods:The cellar model of AD was eatablished by action of microglial conditioned medium(MCM) induced by aggregated Aβ1-40 on rat cultured hippocampal neurons.We added triotolide in different dose to MCM,then divided the hippocampal neurons into four groups:Model group,Low-dose TP-MCM group , High-dose TP-MCM roup and Normal-MCM group.After 2 hours or 24 hours of giving drugs, MTT assay and TUNEL staining were used to observe the effects of triptolide on the neuronal apoptosis in hippocampus of the cellular model of AD , Normal group which was added nothings is the Control group.Results:After 2 hours of giving MCM,the apoptotic number of hippocampal neurons in every group have no significant difference.After 24 hours of giving MCM,the apoptotic number of hippocampal neurons in Model-MCM group was more higher than other groups.Compared with low-dose TP-MCM group, the apoptotic number of hippocampal neurons in High-dose TP-MCM group was obviously decreased. The apoptotic number of hippocampal neurons of in High-dose TP-MCM group,Normal-MCM group and Normal group have no significant difference.Conclusions:Triptolide could restrain the neuronal apoptosis in hippocampus of the cellular model of AD.
