| Objective:To detect the expression of macrophage migration inhibitory factor (MIF) in the retina of diabetic rats, to investigate its role in the pathogenesis of diabetic retinopathy(DR), the effect and significanc of curcumin early intervention on expression of MIF in retina of diabetic rats.Methods:1,Diabetic rats were induced by intraperitoneal injection of STZ , and the diabetic rats were randomly allocated in to diabetic group and curcumin group , also set the normal control group , N=10 .2,Give the treatment group rats with curcumin 200mg/kg·d-1 by intragastric administration , with 1% carboxymethyl cellulose suspension before the adiministration . At the same time the control group and diabetic group were accepted the same amount of carboxymethyl cellulose , last for 8 weeks before the end of experiment , and motitor blood glucose and weight of rats in all experimental groups at 1week,2weeks,4weeks and 8weeks.3,Detect the different content of MIF in retina of every group by ELISA , the content of MIF of every group were statistically analyzed , P<0.05 was considered statistically significant.4,HE staining and immunohistochemical staining were used to observe the expression and localization of MIF in retina , electron microscope was used to observe the ultrastructural changes in the retina .Results:1,In the experiment period, blood glucose of diabetic group and curcumin group was signifcantly higher than normal control group(P<0.05). Compared with diabetic group , blood glucose of curcumin group decreased after 2weeks(P<0.05). The weights of diabetic group decresed compared with normal control group(P<0.05). The weights of curcumin group was lower than normal control group(P<0.05), and higher than diabetic group after 2weeks(P<0.05).2,The expression of MIF in retina of dibetic group and curcumin group was signifcantly higher than normal control group(P<0.01). Curcumin group reduced the expression of MIF in retina compared with dibetic group(P<0.01), the difference has statistically significant.3,Immunohistochemical staining showed diabetic model rats had a lot of MIF expressed in the retina , mainly expressed in the inner nuclear layer, outer nuclear layer and retinal capillary endothelial cells . Normal control group rats did not express or had a small amount of expression in retina . Expression of MIF in retina of curcumin group was significantly reduced compared with diabetic group .4,Histological observation showed that the retina of diabetic rats had corresponding changes with early diabetic retinopathy , the retina changes of curcumin group rats were slighter than diabetic group .Conclusion :1,The expression of MIF in retina of diabetic rats increased, related to the retinal inflammtory change , may participate in early diabetic retinopathy .2,Curcumin can effectively down-regulate the expression of MIF in retina , its early intervention can delay the disease process . |
