Objective:To explore the relationship of the expression of HPV16-E6,HPV16-E7,Survivin in cervical intraepithelial neoplasia and squamous cell carcinoma of cervix and the malignant progression of cervical precancerous lesions. Methods:The expression of HPV16-E6,HPV16-E7 proteins and survivin in 30 cases of squamous cell carcinoma of cervix (SCC), 90 cases of cervical intraepithelial neoplasia(CIN, including CIN1, CIN2, CIN3, respectively 30 cases) and 10 cases of normal cervical tissues were detected using immunohistochemical Elivision method. Firstly, Kruskal-Wallis H test was used to compare the overall distribution of expression of the detection factors, then, Wilcoxon rank sum test was used to compare the factors distribution in each group. Spearman rank correlation was used to analyze the relationship among the three factors. Results:In control group, CINⅠ,CINⅡ,CINⅢ,SCC, the expression of HPV16-E6 was 20.0%, 30.0%, 40.0%, 70.0%, 86.7%,the expression of HPV16-E7 was 30.0%, 63.3%, 63.3%, 76.7%,96.7%,the expression of Survivin was 60.0%, 76.7%, 86.7%,93.3%,93.3%。The positive degree of the expressions of the three factors all showed increasing trend with the progression of the cervical lesion. The expressions of HPV16-E6 and HPV16-E7 in CIN3 and SCC were all significantly higher than that of normal cervical tissues(P<0.05). The expressions of Survivin in CIN2, CIN3 and SCC were significantly higher than that of normal cervical tissues(P<0.05).Correlation: In the progression of cervical lesions, HPV16-E6 and HPV16-E7 expressions were positively correlated (rs=0.356,P<0.001), HPV16-E6 and Survivin expressions were positively correlated (rs=0.322,P<0.001), HPV16-E7 and Survivin expressions were positively correlated (rs=0.302,P=0.001). Conclusion:1. The positive expression degree of HPV16-E6, HPV16-E7 and Survivin was gradually increasing with the progress of cervical lesions.2. In the procedure of the progression of CIN, HPV16-E6, HPV16-E7, and Survivin were all positively correlated.3. HPV16-E6, HPV16-E7 and Survivin could be considered as prognostic factors of cervical intraepithelial neoplasia.
