| Objective:To analyze Calcium signal transduction mechanism in the role of diacetylmorphine arrhythmogenic.These experiment concloud rats mental and physical dependence in animal behavior;To explore various types of arrhythmia by diacetylmorphine until rats death,and to explore the effect on membrane potentials and L-type calcium channel currents in arrhythmia rats After the diacetylmorphine. Methods:We are use the method of the human abuse diacetylmorphine to building the addicted rats, used the steadily increase way about diacetyl of morphine dosage, Our experiment used the steadily increase way about diacetyl of morphine dosage,the experiment rats were Wister rats, First dosage was 5 mg/(kg?d),three times everyday,increased 2.5 mg/kg everyday,Observed the symptom of withdraw after injected naloxin in order to judge whether the model success.When the experiment rats were abused,we would continue add the diacetyl of morphine dosage in order to establish the experiment rats proarrhythmia model,when the rats dead,we would analysis the rats proarrhythmia types.when the proarrhythmia appeared,we would detect the change of the cell membrane potential and L-type calcium channel current changes by the technology of whole-cell patch clamp.Results:1) We were contrasting the experimental group and control group during the experiment,the rats displayed a variety of spiritual and physical behaviors of drug dependence.2) When the experiment rats were abused by diacetyl of morphine,We would find a series of animal behaviors,and when the rats proarrhythmia appeared then we found all kinds of proarrhythmia about rats death.3) The I-V curves were shifted upward of the rats L-type calcium electric current,the peak currents clearly reduced contrast to control groups,they were significantly diferent(P<0.05),the APD90 curtailed evidently in the action potentia. Conclusion: Diacetyl of morphine could cause the rat cardiomyocytes L-type calcium electric curren reduced,and it could lead to the rats show proarrhythmia.We could clear the interrelated system were damaged by diacetyl of morphine.these research findings could provide basis theories for clinical prevention and legal medicine.We could receive all kinds of proarrhythmia by these bused rats,these research findings could provide basis theories for clinical treatment. |
