Objective Use the Immunohistochemical method (SABC) to detect the different expressions of PTEN and PI3K in superficial gastritis, atrophic gastritis, with intestinal metaplasia, dysplasia and gastric cancer tissue .The effects of PI3K-p110βin the occurrence and evelopment and clinical significance of gastric cancer offer theoretical basis for the diagnosis, treatment and prognosis.Method First,Select Yan'an University Hospital Pathology as the place to do the research which has plenty of clinical and pathological data,including gastroscopy and surgical biopsy specimens simple between2008 and 2009: superficial gastritis 19 cases; simple atrophic gastritis 11 cases; superficial with intestinal metaplasia 5 cases atrophy with intestinal metaplasia 14 cases; superficial with mild dysplasia 9 cases, moderate dysplasia 5 cases severe dysplasia 6 cases; atrophy with mild dysplasia 11 cases, moderate dysplasia 3 cases severe dysplasia 3 cases; well differentiated gastric adenocarcinoma 13 cases differentiated gastric adenocarcinoma 11 cases, poorly differentiated gastric adenocarcinoma 17 cases, intramucosal carcinoma 2 cases. Those were detected by using SABC gastric mucosa the expression of PTEN and PI3K situation.Second,the data were analyzed for significance using the statistical software SPSS 17.0 computer package.Result (1) PTEN protein was located in cytoplasm of gastric mucosa, chronic gastritis in the positive rate of 94%, dysplasia 88.9%, gastric cancer was, 74.4%, the difference was significant (P = 0.000 <0.05). (2) PTEN protein's expression had nothing to do with the patients'age and gender but was related to the pathogenesis's location (P = 0.042 <0.05).(3) PI3K protein was located in cytoplasm of gastric mucosa, chronic gastritis in the positive rate of 20%, dysplasia 63.9%, gastric cancer was 100%, the difference was significant (P = 0.000 <0.05).(4) PI3K protein's expression had no relation with the patient's age and gender.,but was related with the pathogenesis's location (P = 0.006 <0.05).(5) PTEN protein in the PI3K protein in gastric tissue stained positive rate (53.2%) was much lower than the PTEN protein-negative gastric mucosa PI3K protein staining positive rate (94.4%) (chi-square value = 10.910, P = 0.001 <0.05); and Spearman correlation analysis and PI3K PTEN protein expression was negatively correlated (r =- 0.643, P = 0.000 <0.05)Conclusion 1, Expression of PTEN in patients,who with chronic gastritis, dysplasia, or gastric cancer, a gradual decline and was closely related with the pathological changes, indicating that the loss expression of PTEN was associated with the development of gastric cancer.2, PI3K-p110βin chronic gastritis, dysplasia, gastric's expression was increasing, gradually,and its expression was closely associated with the lesions, indicating that PI3K-p110βexpression was associated with the development of gastric cancer.3, Comparing the expression of PI3K-p110βin absence of PTEN in gastric cancer with the expression of PTEN in gastric cancer ,the former was significantly increased, and this suggested that PTEN's loss in gastric cancer, the tumor growth and proliferation was dependent on p110βsignaling effect.4, PTEN's and PI3K-p110β's expressions were related to the differert location of gastric cancer, suggesting that non-antral gastric cancer and gastric antrum in the pathogenesis of gastric cancer may be different.
