Objective:This experiment study antibacterial effect of ShuangHuangLian powder injection, Shuanghuanglian powder injection combined with cefoperazone/ sulbactam on extensively drug resistant acinetobacter baumannii in vitro, to validate antibacterial effect of Shuanghuanglian to extensively drug resistant acinetobacter baumannii. This experiment discuss antibacterial effect of combined medicine on extensively drug resistant acinetobacter baumannii, and apply for the experimental basis for clinical treatment of extensively drug resistant acinetobacter baumannii infection. This experiment intervene on Shuanghuanglian powder injection on resistant strains of acinetobacter baumannii to reduce and delay the extensively drug resistant strains of acinetobacter baumannii, which lay the foundation for antibacterial mechanism of ShuangHuangLian powder injection and traditional Chinese medicine in vivo and clinical research.Method:This experiment isolate from my hospital clinical sputum, to get pure bacterial colonies detemined by automatic microbial identification and susceptibility analysis system (Phenix-100), antimicrobial susceptibility follow CLSI M100-S24, to electe extensively drug resistant Acinetobacter baumannii. This experiment use the broth dilution method with different concentrations of ShuangHuangLian powder injection and ShuangHuangLian powder injection+cefoperazone/sulbactam combination in 35 ℃ for 24 hours, to determine the minimal inhibitory concentration.Result:The minimal inhibitory concentration(MIC) of ShuangHuangLian for extensively drug resistant acinetobacter baumannii is 20mg/mL, the minimal inhibitory concentration(MIC) of Cefoperazone/sulbactam is 256ug/mL. When Shuanghuanglian combined with cefoperazone/sulbactam, Shuanghuanglian concentration is 7.5mg/mL, cefoperazone/sulbactam concentration of 64ug/mL inhibited the growth of extensively drug resistant acinetobacter baumannii; Shuanghuanglian concentration is 10mg/mL, then cefoperazone/sulbactam concentration of 32ug/mL inhibited the growth of extensively drug resistant acinetobacter baumannii, cefoperazone/sulbactam concentration is reduced by 8 times combined than alone. Shuanghuanglian and cefoperazone/sulbactam combined effects of extensively drug resistant acinetobacter baumannii enhance the antibacterial effect than Shuanghuanglian or cefoperazone/sulbactam alone.Shuanghuanglian combined with cefoperazone/sulbactam simultaneously apply on extensively drug resistant acinetobacter baumannii and delayed 2 hours, delayed 4 hours, delayed 6 hours apply on extensively drug resistant acinetobacter baumannii. the effect showed:When Shuanghuanglian same concentration, the cefoperazone/sulbactam concentration of each delayed hours group were compared with the simultaneous group is the same or lower, cefoperazone/sulbactam concentration of delayed 4 hours group is lower than the other groups, delayed 4 hours group antibacterial effect is better than other groups.Conclusion:The experimental results show that ShuangHuangLian powder injection has certain antibacterial effect of extensively drug resistant acinetobacter baumannii, Shuanghuanglian powder injection and Cefoperazone/sulbactam combined effects of extensively drug resistant acinetobacter baumannii, greatly enhance the antibacterial effect than the drug alone,and Shuanghuanglian powder injection combined with cefoperazone/sulbactam delayed 4 hours group is best.The application of Shuanghuanglian powder injection combined with cefoperazone/sulbactam can reduce the amount of antibiotics, to reduce and delay the bacterial resistance.
